摘要
目的探讨应激性心肌病模型大鼠心肌组织中溶酶体组织蛋白酶L(cathepsin L)和葡萄糖调节蛋白78(GRP78)表达变化,及其在应激性心肌病法医学诊断中的意义。方法将25只大鼠随机分为模型组(15只)和对照组(10只)。模型组大鼠采用束缚加电击的复合应激方法造模,对照组不做任何处理。应用放射免疫法检测两组大鼠血清皮质醇的含量,免疫组化方法检测大鼠心肌组织cathepsin L和GRP78蛋白的表达。结果模型组大鼠血清皮质醇含量明显高于对照组(t=8.78,P<0.05),造模成功。模型组大鼠心肌组织cathepsin L和GRP78蛋白表达量均高于对照组,差异有统计学意义(t=10.44、2.55,P<0.05)。结论应激性心肌病大鼠心功能受损,cathepsin L和GRP78表达升高,二者检测有望成为法医学诊断应激性心肌病所致心源性猝死的重要辅助手段。
Objective To explore the expressions of lysosomal cathepsin L and glucose-regulated protein 78(GRP78)in cardiac muscle of a rat model of irritability myocardial disease(IMD)and its significance in forensic diagnosis of IMD. Methods Twenty-five rats were randomized to model group(n=15)and control group(n=10).A model of IMD in rats was created in the model group using combined stress of restraint and electric shock.No any processing was done for those in the control group.Employing radioimmunoassay method to detect serum levels of cortisol of rats in both groups,and immnohistochemical technique to detect the expressions of cathepsin L and GRP78 in myocardial tissue. Results The level of serum cortisol in rats of the model group was much higher than that in the control group(t=8.78,P〈0.05),explaining the success of modelling.The expressions of protein cathepsin L and GRP78 were significantly higher than that in the control group(t=10.44,2.55;P〈0.05). Conclusion Cardiac function is damaged in rats with irritability myocardial disease.The expressions of cathepsin L and CRP78 elevate,a detection of both markers is expected to become an important assisting means in forensic diagnosis of sudden cardiac death due to irritability myocardial disease.
