摘要
目的探索脂多糖的作用下单核/巨噬细胞产生的炎症因子与急性冠脉综合征病程的内在关联。方法体外培养人急性单核细胞白血病细胞(THP-1),实验组加脂多糖,对照组空白,按一定的时间间隔取一定体积培养液,用ELISA检测IL-1β、IL-6、IL-1α浓度,观测其变化情况及何时达到峰值。并对出现峰值最早的IL-1β进行实时定量PCR检测。结果 ELISA显示,随着培养时间的延长,浓度也发生相应的改变,IL-1β在3时达到峰值,IL-6在16时达到峰值,IL-1α在18时达到峰值。实验组与对照组比较差异有统计学意义(P<0.05)。结论脂多糖影响巨噬细胞IL-1β、IL-6、IL-1α的表达;达到峰值的时间不同,说明IL-1α、IL-1β、IL-6可能在不同的时间起主导作用。
Objective To explore the relationships between acute coronary syndrome and inflammatory fac-tors which were produced by macrophages with lipopolysaccharide (LPS). Methods The THP-1 cells were cultured in vitro and divided into research group and control group at random. LPS was used in the research group, but not in the control group. The concentration of interleukin-1β(IL-1β), interleukin-1α(IL-1α), interleukin-6 (IL-6) were de-termined by ELISA. The expression of IL-1β was determined by real time PCR. Results The results of ELISA showed that the concentrations of IL-1β, IL-1α, IL-6 were changing with time courses. The peak value of IL-1βwas observed at 3 h, with that IL-6 observed at 16 h and that of IL-1αobserved at 18 h. The differences between the re-search group and the control group were statistically significant (P〈0.05). Conclusion The LPS affects the expres-sion of IL-1β, IL-6, IL-1αof macrophages, with the peak values observed at different time points, which indicates that IL-1β, IL-6 and IL-1αplay the leading role at different times in the process of acute coronary syndrome.
