摘要
目的 探讨小鼠左冠状动脉前降支结扎导致急性心肌梗死时小鼠血清心肌肌钙蛋白I(cardiac troponin I,cTnI)的变化及其与心肌梗死面积的相关性。方法 8~12周龄雄性C57BL/6小鼠21只,分为假手术组、低位结扎组和高位结扎组各7只,低位结扎组和高位结扎组分别在冠状动脉左前降支低位和高位实施结扎术,假手术组手术步骤同前,但未行左冠状动脉前降支结扎。3组术后2、6、12h采用ELISA法检测血清cTnI水平,术后12h处死3组小鼠行组织病理学检查测量心肌梗死面积。结果 术后6、12h,低位结扎组小鼠血清cTnI水平[(31.84±4.52)、(105.03±23.43)μg/L]、高位结扎组[(35.18±7.47)、(132.06±40.95)μg/L]高于假手术组[(24.91±3.79)、(65.28±5.76)μg/L](P〈0.05),高位结扎组高于低位结扎组(P〈0.05);高位结扎组小鼠心肌梗死面积[(58.50±6.24)%]高于假手术组[(1.77±0.60)%]和低位结扎组[(38.88±7.97)%],低位结扎组高于假手术组(P〈0.05);术后2、6、12h血清cTnI水平与心肌梗死面积均呈正相关(r=0.452,P=0.009;r=0.662,P=0.001;r=0.713,P=0.000)。结论 急性心肌梗死后小鼠血清cTnI水平快速升高,且cTnI水平与心肌梗死面积呈正相关,心肌梗死12h内,随时间延长,cTnI与心肌梗死面积的相关度逐渐增高。
Objective To evaluate the changes of serum cardiac troponin I (cTnI) after acute myocardial infarction (AMI) induced by ligating the anterior descending branch of left coronary artery, as well as to analyze the its correlation with myocardial infarct size. Methods Twenty-one wild-type C57BL/6 male mice aged from 8 to 12 weeks were divided into three groups: sham-operation group, low-ligation group and high-ligation group, with 7 mice in each group. The anterior descending branch of left coronary artery was ligated in low-ligation group and high-ligation group, and sham- group was performed the same procedures except the final ligation. The serum eTnI level was measured by ELISA method 2, 6 and 12 hours after AMI. All animals were sacrificed 12 hours after AMI, and the myocardial infarct size was calculated by histopathologic method. Results After AMI, the cTnI levels were the highest 6 and 12 hours after AMI in high-ligation group ((35.18±.47), (132.06±40.95) μg/L), followed by those in low-ligation group ((31.84±4.52), (105.03±23.43) μg/L) and sham operation group ((24.91±3.79), (65.28±5.76) μg/L) in turns (P〈0.05). The myocardial infarct size was the largest in high ligation group ((58.50±6.24)%), followed by that in low-ligation group ((38.88±7.97)%) and sham-operation group ((1. 77±0. 60)%) in turns (P〈0.05). The cTnI levels 2, 6 and 12 hours after AMI were positively correlated with myocardial infarct size (r=0. 452, P=0. 009; r=0. 662, P=0. 001; r= 0. 713, P=0. 000). Conclusion The serum cTnI level increases rapidly, and is positively correlated with myocardial infarct size. In 12 hours after AMI, the correlation degree between cTnI and myocardial infarct size increases gradually with the prolong of time.
