α-Hydroxyl carboxylic acids and their derivatives exist as the key substructures in a variety of biologically active natural products and numerous pharmaceuticals. Various -COX synthons(X=OH, or the equivalent), which are useful umpolung agents, have been developed. Recently, Nemoto et al. prepared the masked acyl cyanides[2, HC(CN)2OR3],α-COX synthon that played an important role in the synthesis of a-hydroxyl carboxylic acids and their derivatives, from commercially available materials in three steps under mild conditions(Scheme 1).
