摘要
利用高效液相色谱建立了粮食中黄曲霉毒素B1的测定方法。样品经乙腈水溶液(乙腈与水体积比为84:16)提取,提取液经多功能净化柱净化、浓缩,碘衍生试剂柱前衍生、C18色谱柱分离,荧光检测器检测,外标法定量。选择线性范围在0~5.0 ng/m L之间,结果表明线性良好,方法的检出限为0.17μg/kg。对添加高、中、低3个浓度黄曲霉毒素B1的大米样品进行加标回收实验,平均回收率为92.2%、91.0%、95.2%,变异系数为2.1%、1.7%、2.3%。
The article is to seek a rapid and reliable detection method by HPLC for AFB1 content measurement in grains.The sample is extracted by acetonitrile and water volume ratio(84:16),purified and concentracted by Myco Sep 226 Afla+Zon column,derived by I2 solution,separated by C18 chromatographic column, detected by fluorescence detector, and then quantitatived by external standard method.The linearity range is between 0 and 5.0ng/mL,and the results shows that it is very good and the detection limit is 0.17 μg/kg. The recovery experiment is carried on three levels, and the average recovery rate is separately 92.2%,91.0%,95.2%, the RSD is separately2.1%,1.7%,2.3%.
