营养物质及激素组合对梭梭无菌苗生长的影响

摘要 以梭梭种子为实验材料,以1/2MS、1/4MS、MS为基本培养基,考察水解酪蛋白(CH)和肌醇(IP)、蔗糖、pH 值、硝酸银及激素组合对梭梭无菌苗腋芽萌发及丛生芽形成的影响.结果表明,不添加水解酪蛋白(CH)和肌醇(IP)的处理,其单株真叶数及相对生长率分别提高了83.3%、1倍;1/4MS+蔗糖10g/L+IBA10.0mg/L 或AgNO3 0.5mg/L+IBA5mg/L+6GBA1.0mg/L、pH5.8-8.0时,单株真叶数的相对生长率最大;苗龄为40d、MS+6GBA0.6mg/L+硝酸银4.0mg/L时块均芽数最多为14个.添加水解酪蛋白(CH)和肌醇(IP)对梭梭无菌苗腋芽萌发没有促进作用,低浓度蔗糖及硝酸银有利于梭梭无菌苗腋芽萌发,较高浓度的硝酸银有利于无菌苗形成丛生芽. Haloxylon ammodendron aseptic seedlings were applied as experimental materials,1/2MS,1/4MS,MS as the basic culture medium to investigate the effects of casein hydolysate(CH)and sucrose,pH and hormone combinations on the germination of axellaxy buds of sterile seedlings Haloxylon ammodendron and formation of adventitious buds of sterile seedlings.The results indicated that when there were no treatments with hydrolyzed protein and inositol,the leaf number of per plant and relative growth rate were respectively increased by 83.3%;when there were 1/4MS+sucrose 10g/L+IBA 10mg/L or AgNO30.5mg/L +IBA 5mg/L +6-BA 1.0mg/L,pH 5.8-8.0,the relative growth rate of the leaf number of per plant was the greatest;when the seedling age was 40 d,MS+6-BA 0.6mg/L+AgNO34.0mg/L,the greatest number was 14.Low concentration of sucrose and AgNO3 were conducive to the germination of axillary buds of sterile seedlings Haloxylon ammodendron.The higher concentration of AgNO3 is conducive to the formation of adventitious buds of sterile seedlings.
