摘要
为了进一步提高节杆菌P-1酶解PVA废水的效率,用单因素实验考察废水初始PVA浓度、酵母汁加入量、HRT、温度对节杆菌P-1的PVA降解酶活性和PVA降解率的影响,用Box-Behnken Design实验设计和响应面法进一步优化上述4因素.单因素得出的最适初始PVA质量浓度、酵母汁加入量、HRT、温度分别为833 mg/L、0.45 g/L、24 h、30℃.响应面法得出的节杆菌P-1的最佳产酶条件为:初始PVA质量浓度680 mg/L、酵母汁加入量0.4 g/L、运行时间22 h、处理温度29℃.在此最佳条件下,PVA降解酶的活性为3.34 U,PVA降解率达到94.12%,与预测值3.23 U非常接近.各因素影响PVA降解酶活性的主次顺序为:运行时间>初始PVA质量浓度>酵母汁加入量>温度.
Single factor experiments were used to investigate the effects of poly( vinyl alcohol)( PVA) concentrations,yeast added,run time and temperature on the PVA ase activity and PVA degradation rate of Arthrobater sp. P- 1. And then Box- Behnken Design and response surface methodology were constructed to further optimize the four factors to improve PVA ase activity. The results indicated that the appropriate PVA concentration,yeast added,run time and temperature determined by single factor tests were 833 mg /L,0. 45 g / L,24 h and 30 ℃,respectively. The optimal conditions to produce PVA ase of Arthrobater sp. P- 1 were 680 mg / L PVA,0. 4 g / L yeast added,22 h and 29 ℃. Under the optimal conditions,the activity of PVA ase and PVA degradation rate were3. 34 U and 94. 12%,respectively. The order of effects on the PVA ase was as follows: run time PVA concentration yeast added temperature.
