铁皮石斛体细胞胚胎发生类受体激酶基因DoSERK的克隆和表达分析 被引量:5

Cloning and Expression Analysis of Somatic Embryogenesis Receptor Kinase Gene(DoSERK) from Dendrobium officinale Kimura et Migo

摘要 为了解铁皮石斛(Dendrobium officinale)中的体细胞胚胎发生类受体激酶基因Do SERK的功能,在转录组测序数据的基础上克隆了Do SERK的全长c DNA。结果表明,Do SERK与其他植物的SERK高度同源,编码633个氨基酸。生物信息学分析表明,Do SERK蛋白为亲水蛋白并定位于质膜,具有1个信号肽、1个富脯氨酸区域、1个跨膜区、5个富亮氨酸重复序列以及1个保守的蛋白激酶活性结构域,属于SERK蛋白家族成员。系统进化树分析表明,Do SERK与同为兰科植物的文心兰(Cyrtochilum loxense)以及卡特兰(Cattleya maxima)的SERK亲缘关系最近。组织表达分析表明,Do SERK在铁皮石斛中广泛表达,以幼嫩小苗根部的表达量最高。这些说明Do SERK除了可能参与铁皮石斛体胚发生过程以外,还参与其他生长发育过程。 In order to understand the function of somatic embryogenesis receptor-like kinase gene(Do SERK) from Dendrobium officinale Kimura et Migo, the full-length c DNA of Do SERK was cloned based on transcription sequences data. The results showed that Do SERK had high homology to SERK gene in other plants, encoding 633 amino acids. Bioinformatics analysis showed that Do SERK was a hydrophilic protein locating in plasma membrane. Do SERK had typical domains of SERK proteins, containing one signal peptide, one specifi c prolinerich region, five leucine-rich repeat motifs, one transmembrane domain and one conservative protein kinase domain. Phylogenetic tree based on SERKs revealed that D. officinale was close to Cyrtochilum loxense and Cattleya maxima of Orchidaceae. Do SERK could widely express in tissues of D. officinale, and the expression was high in roots of seedlings. So, it was suggested that Do SERK not only involved in somatic embryogenesis, but also in many other processes of growth and development.
