摘要
目的 探讨增龄影响下大鼠血管平滑肌细胞(VSMC)前胶原α-多肽的改变及潜在的可能机制.方法 通过对刚出生的幼龄(对照组)和9个月成年(成年组)健康雄性Wistar大鼠的胸腹主动脉进行体外试验,采用RT-PCR半定量和定量技术检测VSMCⅠ型和Ⅲ型前胶原α-多肽mRNA,通过VG染色、Western blot及ELISA方法检测前胶原α-多肽蛋白表达.结果 RT-PCR半定量结果显示,Ⅰ型前胶原α-多肽mRNA对照组为76.62±1.05,成年组为78.37±2.42,两组比较未见统计学差异(P>0.05);Ⅲ型前胶原α--多肽mRNA对照组为105.4±2.66,成年组为123.1±3.81,两组比较未见统计学差异(P>0.05).Ⅰ型前胶原α-多肽mRNA定量成年组为4.63±1.03,但与对照组3.13±0.54比较未见统计学差异(P=0.05);Ⅲ型前胶原α-多肽mRNA定量成年组为7.68±0.63,与对照组6.86±0.41比较未见统计学差异(P>0.05).Ⅰ型前胶原α-多肽蛋白表达在成年组为0.1 ±0.03,对照组为0.06土0.03,两组比较差异有统计学意义(P<0.05);Ⅲ型前胶原α-多肽蛋白表达在成年组为0.58±0.06,对照组为0.4±0.02,差异有统计学意义(P<0.05).结论 前胶原α-多肽增龄性变化的调节通路是老化性血管重构的分子机制之一.
Objective To investigate the age-related changes of proeollagen alpha polypeptide gene mR- NA and protein expression in rat vascular smooth muscle cell (VSMC) and the possible underlying mechanisms. Methods For in vitro culture of VSMC from thoracoabdominal aorta in neonate and 9 months old healthy male Wistar rats, proeollagen alpha polypeptide mRNA and procollagen alpha polypeptide protein expression were de- tected, using of RT-PCR, VG staining, Western blot and ELISA methods. Results Real-time polymerase chain reaction (RT-PCR) Semi quantitative displays that there was no significant difference between the two groups of type Ⅰ and type Ⅲ collagen α polypeptide chain mRNA. RT-PCR quantitative result displays that type Ⅰ colla- gen ot polypeptide chain mRNA increased in adult group, but no significant difference(P=0.05 ), there was no sig- nificant difference between the two groups of type Ⅲ collagen α polypeptide chain mRNA (P〉0.05). Both type Ⅰ and type Ⅲ procollagen alpha polypeptide protein expression were increased significantly in adult group as com- pared with the young group (P〈0.05). Conclusion Regulatory pathway on age-related changes of proeollagen alpha polypeptide is one of the molecular mechanisms of aging vascular remodeling.
