脑心通胶囊对慢性低灌注血管性痴呆小鼠神经保护作用研究 被引量:5

摘要 目的研究脑心通胶囊对慢性低灌注损伤所致的血管性痴呆小鼠模型的神经保护作用。方法取54只C57BL/6小鼠行双侧颈总动脉缩窄术建立慢性低灌注血管性痴呆模型,将造模成功的45只小鼠随机分为模型组(n=15)、Tempol组(n=15)、脑心通组(n=15)。另设假手术组(n=10)。1 d后,Tempol组灌胃给予10 mg/m L Tempol溶液0.01 m L/(g·d),脑心通组灌胃给予脑心通350 mg/(kg·d),假手术组、模型组灌胃给予生理盐水0.01 m L/(g·d),连续给药28 d。采用Y迷宫检测脑心通胶囊对血管性痴呆小鼠学习记忆能力的影响;利用苏木精-伊红(HE)染色和免疫组织化学法检测脑心通胶囊对海马CA1区组织学变化及星形胶质细胞标志物胶质细胞原纤维酸性蛋白(GFAP)的表达情况。结果连续给药28 d后,与模型组[(53.53±19.63)%]比较,Tempol组、脑心通组进臂的自发交替百分比[(65.34±12.32)%、(65.19±4.85)%]明显增加(P<0.05)。HE染色显示模型组C57BL/6小鼠的海马CA1区锥体细胞排列紊乱、稀疏,细胞周围间隙增大,细胞内出现空腔。Tempol组、脑心通组的锥体细胞空腔减小,形态基本接近正常,细胞周围间隙减小,接近正常形态。与假手术组[(27.01±6.18)个/mm2]比较,模型组海马区每平方毫米活化的星形胶质细胞数目[(49.93±4.67)个/mm2]显著增加(P<0.05);与模型组比较,Tempol组、脑心通组的GFAP阳性细胞数目[(38.71±11.67)、(40.68±8.57)个/mm2]明显减少(P<0.05),结构明显改善,分枝少,胞体形态趋于正常。结论脑心通胶囊可明显提高慢性低灌注血管性痴呆小鼠的空间学习记忆能力,改善神经组织形态,同时抑制激活的星形胶质细胞数量,表明脑心通胶囊可以改善血管性痴呆C57BL/6小鼠脑组织的缺血性损伤。 Objective To study the neuroprotective effect of Naoxintong Capsules on vascular dementia mice caused by chronic hypoperfusion damage. Methods Fifty four C57BL/6 mice were taken double carotid artery coarctation operation to establish vascular dementia mice model with chronic hypoperfusion. Forty five successful modeling mice were randomly divided into model group(n = 15), Tempol group(n = 15) and Naoxintong group(n = 15). Ten mice were set as sham group. One day later, Tempol group was given orally administration of 10 mg/m L Tempol solution 0.01 m L/(g·d),Naoxintong group was given orally administration of Naoxintong 350 mg /(kg·d), sham group and model group were given orally administration of normal saline 0.01 m L/(g·d), continued for 28 d. Y maze was used to detect the learning and memory ability of Naoxintong Capsules for vascular dementia mice. The hematoxylin-eosin(HE) staining and immunohistochemistry were used to detect the histological change of Naoxintong Capsules for hippocampus CA1 region and the expression of Naoxintong Capsules for the marker glial filament acidic protein(GFAP) of astrocyte.Results After successive administration for 28 d, compared with model group [(53.53 ±19.63)% ], the spontaneous alternation percentages of entering arm in Tempol group and Naoxintong group [(65.34 ±12.32)%,(65.19 ±4.85)% ] were in creased significantly(P〈0.05). HE staining showed that pyramidal cells of hippocampus CA1 region of C57BL/6 mice in model group were arranged in disorder and dispersely,the peripheral space was enlarged, there was cavity in cells. The cavity of pyramidal cells in Tempol group and Naoxintong group was reduced, the cell morphology was almost normal, the peripheral space was reduced and got close to normal.
