摘要
目的研究β-淀粉样蛋白(Aβ25-35)对大鼠海马细胞内质网钙库的影响。方法将40只成年清洁级雄性SD大鼠按体重随机分为4组,分别为Aβ25-35染毒组(每测一次性注射2.5μg/μl凝聚态Aβ25-35溶液2μl)、假手术组、生理盐水组,正常对照组(不进行任何处理),每组10只。于手术后14 d,检测海马细胞内游离钙离子浓度和内质网IP3、RYR、PS mRNA的表达水平。结果与正常对照组、假手术组和生理盐水组相比,Aβ25-35染毒组海马细胞内质网肿胀肥大,扩张明显,且大多内质网膜破裂,游离钙浓度明显增高,内质网IP3、PS mRNA的表达水平升高,RYR mRNA的表达水平降低,差异有统计学意义(P<0.05)。结论 Aβ25-35可以通过破坏大鼠海马细胞内质网钙稳态而发挥神经毒性作用。
Objective To study the effects of β-amyloid(Aβ25-35) on endoplasmic reticulum calcium stores in hippocampal cells of rats. Methods A total of 40 adult male SD rats were randomly divided into four groups by weight, A β25-35 exposure group(each side injection 2 μl 2.5 μg/μl condensed Aβ25-35), sham operation group, normal saline group and normal control group(no treatment), ten in each group. At the 14 th day after surgery, the concentration of intracellular free Ca2 +and the expression levels of IP3, RYR, PS gene m RNA in hippocampus of rats were detected respectively. Results Compared with the normal control group, the normal saline group and the sham operation group, endoplasmic reticulum swelled and showed hypertrophy,expanded obviously, and most endoplasm retinal bursted in hippocampal cells of rats exposed to A β25-35,the levels of intracellular free Ca2+increased in hippocampus cells of rats exposed to Aβ25-35, IP3 and PS m RNA expression increased, RYR m RNA expression reduced in endoplasmic reticulum of hippocampus cells of rats exposed to A β25-35, and revealed statistically significant difference(P〈0.05). Conclusion Aβ25-35 can play a significant role of neurotoxicity by destroying calcium homeostasis of endoplasmic reticulum in hippocampal cells of rats.
