川牛膝在去卵巢大鼠体内的雌激素效应及对脂代谢的影响 被引量:11

Estrogen effect of Cyathnla capitata Moq. in ovariectomized rats in vivo and its impact on lipid metabolism

摘要 目的观察川牛膝在去卵巢大鼠体内的雌激素效应及对脂代谢的影响。方法选取3月龄雌性SD大鼠50只随机分为5组:假手术组(SHAM组)、单纯卵巢切除组(OVX组)、雌激素对照组(ERT组)、高剂量川牛膝组(CH组)和低剂量川牛膝组(CL组)。除SHAM组外各组均切除卵巢,术后1周开始给药,给药期间观察阴道脱落细胞涂片及动情周期改变,连续12周。处死大鼠后检测血脂4项,测量子宫重量,病理切片观察子宫内膜及腺体变化,术前及处死前分别称重,计算体重变化。结果与SHAM组比较,OVX组大鼠阴道上皮细胞角化率和子宫系数均明显降低(P<0.05),子宫内膜及腺体萎缩,血HDL-C降低(P<0.05),LDL、TC、TG升高(χ2<0.05)。川牛膝能使去卵巢大鼠阴道上皮细胞角化率、子宫系数及腺腔增加(P<0.05),子宫内膜增厚,血清HDL-C升高(P<0.05),LDL、TC、TG降低(P<0.05)。结论川牛膝在去卵巢大鼠体内具有弱雌激素样作用,能减轻雌激素缺乏导致的生殖系统萎缩,改善血脂代谢,减轻体重,对治疗更年期综合征可能有效。 Objective To observe the estrogen effect of Cyathnla capitata Moq. in ovariectomized rats in vivo and its impact on lip- id metabolism. Methods Fifty female SD rats aged three months old were selected and randomly divided into sham- operation group (SHAM group), ovariectomized group (OVX group), estrogen control group (ERT group), high-dose Cyathnla capitata Moq. group (CH group) , and low-dose Cyathnla capitata Moq. group (CL group) . Except the rats in SHAM group, the rats in the other four groups under- went ovariectomy; the rats were treated after one week of operation, the smears of vaginal exfoliated cells and changes of estrous cycle were observed for 12 weeks. Four blood lipid parameters were detected after executing the rats, uterine weights were measured, the changes of en- dometrium and glands were observed by pathological smears, the rats were weighted before operation and after executed, the changes of body weight were calculated. Results Compared with SHAM group, the cornification rate of vaginal epithelial cells and uterine coefficient in OVX group decreased significantly (P〈0. 05 ) , endometrium and glands atrophied, blood high-density lipoprotein cholesterol level decreased ( P〈0. 05 ), the levels of low-density lipoprotein cholesterol, total cholesterol ( TC ), and triglyceride (TG) increased significantly (x^2 〈 0. 05 ) . Cyathnla capitata Moq. improved the eornification rate of vaginal epithelial cells, uterine coefficient, and glandular cavity of ovari- ectomized rats (P〈0.05) , endometrium thickened, blood HDL-C level increased (P〈0.05), the levels of LDL, TC, and TG decreased ( P〈0. 05 ) . Conclusion Cyathnla capitata Moq. plays a weak estrogen-like role in ovariectomized rats in vivo, which can relieve atrophy of genital system induced by estrogen deficiency, improve blood lipid metabolism, and reduce body weight; Cyathnla capitata Moq. may be effective for treatment of menopausal syndrome.
