摘要
【目的】观察丹参酮ⅡA对胃癌细胞增殖的影响及其与环氧合酶-2(COX-2)表达的关系。【方法】体外培养胃癌细胞株SGC7901,经丹参酮ⅡA作用后,采用四唑盐(MTT)比色法检测细胞增殖,流式细胞仪检测细胞凋亡,免疫细胞化学sABC法检测COX-2蛋白表达,放射免疫法检测前列腺素E2(PGE2)含量。【结果】丹参酮ⅡA对胃癌细胞的生长有明显的抑制作用,并呈剂量依赖性。以0.5mg/L作用终浓度抑制作用最明显,其48h的抑制率为71.1%,与对照组相比差异有显著性(P〈0.01);5mg/L丹参酮ⅡA作用后,随时间的延长,凋亡率逐渐升高,48h达到高峰,随后逐渐下降,与对照组比较,各处理组都有显著性差异(P〈0.05);丹参酮作用组胃癌细胞COX-2表达明显减少,其培养液中PGE2的产生量下降,与对照组相比差异均有显著性(P〈0.01)。【结论】丹参酮ⅡA可能是通过下调COX-2mRNA的表达水平发挥其对胃癌细胞生长抑制及促进凋亡作用。
[ObiectivelTo observe the effect of TanshinonellA on proliferation and apoptosis of human gastric carci noma cell line SGC7901 and its correlation with COX-2 expression.[Methods]Human gastric carcinoma cell line SGC7901 was cultured in vitro and treated with Tanshinone HA. The cell proliferation was detected by MTT assay, and the cell ap- optosis was detected by flow cytometry, the expression of COX-2 protein was detected by immunohistochemistry SABC method, and the content of prostaglandin E2 (PGE2) was detected by radioimmunoassay.[Results]The growth of gastric cancer cells was significantly inhibited by TanshinoneⅡA , and dose dependence there occurred. The inhibition effect of 0.5 mg/L was the most obvious dosage, and the inhibition rate of 48 h reached 71.1%, the difference was significant com- pared with the control group ( P 〈0.01) ; With 5 mg/L of TanshinonelIA trestment , the apoptosis rate increased grad ually with the time and reached its peak at 48th h, then gradually decreased. Compared with the control group, the treat- ment group had significant difference ( P 〈0.05) ; COX-2 expression in gastric cancer cells of the treatment group was significantly decreased, and the production of PGE2 in the culture fluid decreased significantly ( P 〈0.01).[Conclusion] TanshinoneUA may play a role in the growth inhibition and apoptosis of gastric cancer cells by down regulating the expres- sion of COX-2 mRNA.
