摘要
为探讨高致病性猪繁殖与呼吸综合征病毒(HP-PRRSV)对Marc-145细胞抗氧化功能的影响及α-硫辛酸对感染病毒后细胞氧化应激的影响,将安全浓度的α-硫辛酸添加到接种HP-PRRSV的Marc-145细胞培养液中,将细胞分为病毒组(D组)、α-硫辛酸干预病毒组(DLA)、α-硫辛酸干预组(LA组)和正常细胞对照组(NC组)。2h后开始检测α-硫辛酸对HP-PRRSV接种Marc-145细胞后细胞内过氧化氢(H2O2)和丙二醛(MDA)含量、超氧化物歧化酶(SOD)和过氧化氢酶(CAT)活性。结果表明,与NC组相比,D组细胞H2O2和MDA含量显著增加(P<0.05),SOD活性显著减小(P<0.05),CAT活性显著增强(P<0.05);α-硫辛酸干预后可改善上述指标(P<0.05)。说明HP-PRRSV能够显著引起细胞氧化应激反应;α-硫辛酸能够抑制氧化应激反应,对Marc-145细胞有保护作用。
To study the effect of Highly pathogenic PRRSV (HP-PRRSV)on antioxidant function of Marc-145 cells and the effect ofα-lipoic acid(LA)on oxidative stress of Marc-145 cells infected with HP-PRRSV.After LA within cellular safety concentration was added to Marc-145 cells infected with HP-PRRSV,the Marc-145 cells were divided into three groups of virus-infected (group D),LA-treated-virus (group DLA)and LA-add(group LA).The normal Marc-145 cells were served as the control(group NC). The levels of hydrogen peroxide (H2 O2 ),malondialdehyde(MDA)and the activity of superoxide dismutase (SOD),catalase(CAT)were detected 2 hours later.Compared with group NC,the levels of H 2 O2 and MDA were significantly elevated (P <0.05),and the activities of SOD and CAT were significantly reduced in group D (P <0.05).All markers above were improved by treatment with LA (P <0.05).HP-PRRSV can cause Marc-145 cell high oxidative stress,and LA has a protective effect on Marc-145 cell through in-hibiting oxidative stress.
