摘要
目的研究载脂蛋白E基因多态性对丹参素调脂作用的影响。方法用逆转录聚合酶链反应法检测不同基因型载脂蛋白E与丹参素共同作用于L-02细胞后,L-02细胞低密度脂蛋白受体和羟甲戊二酰辅酶A还原酶mRNA表达的变化。结果Apo E能够明显减弱丹参素对LDLR mRNA表达的上调(P<0.01),突变型E2(Arg112Cys)减弱强度明显大于野生型E3和突变型E4(Cys 158Arg,P<0.01);Apo E亦能够增强丹参素对羟甲戊二酰辅酶A还原酶mRNA表达的下调,E4增强作用最大(P<0.01),但E2、E3作用不明显(P>0.05)。结论载脂蛋白E 112和158位均可能是影响丹参素调脂作用的关键位点。
Objective To study the action of apolipoprotein E genetic polymorphism on the efficacy of Tanshinol. Methods Through detecting the expression of low density lipoprotein receptor mRNA and 3- hydroxy- 3- methylglutaryl coenzyme A reductase mRNA in L- 02 cells by reverse transcription- polymerase chain reaction,to compare the effect of different genetic subtypes of Apo E on Tanshinol. Results Apolipoprotein E can weaken the induction of low density lipoprotein receptor expression caused by Tanshinol. The weakening extent of apolipoprotein E2( Arg112Cys) was stronger than apolipoprotein E3 and apolipoprotein E4( Cys 158Arg)( P〈 0. 01). Apolipoprotein E also can enhance the inhibition of 3- hydroxy- 3- methylglutaryl coenzyme A reductase expression,especially apolipoprotein E4( P〈 0. 01). But apolipoprotein E2 or apolipoprotein E3 can not have statistical significance( P〉 0. 05).Conclusion Both 112 and 158 site might be the key site that have effect on the efficacy of Tanshinol.
