摘要
目的:探讨电针"神门""三阴交"治疗失眠症的作用机制。方法:30只SD大鼠随机分为空白组、模型组、电针组,每组10只,模型组、电针组采用限制性制动法建立失眠模型。造模后,空白组和模型组同步抓取固定但不进行治疗,电针组电针"神门""三阴交"治疗,每天1次,每次15min,治疗4d。治疗后检测各组大鼠昼夜活动量、高架十字迷宫试验进入开放臂百分数和开放臂停留时间百分数,采用酶联免疫法检测各组大鼠血浆去甲肾上腺素(noradrenaline,NE)、多巴胺(dopamine,DA)、肾上腺素(epinephrine,EPI)和丘脑、脑干中NE、DA含量。结果:模型组较空白组白昼活动量明显增多(P<0.05),夜晚活动量显著减少(P<0.05),高架十字迷宫试验进入开放臂百分数和开放臂停留时间百分数均较空白组明显减小(均P<0.05),模型组血浆NE、DA、EPI以及丘脑和脑干中NE、DA含量均较空白组明显升高(均P<0.05);电针组较模型组白昼活动量明显减少(P<0.05),夜晚活动量显著增多(P<0.05),电针组进入开放臂百分数和开放臂停留时间百分数较模型组明显增大(均P<0.05),血浆NE、DA、EPI以及丘脑和脑干中NE、DA含量均较模型组明显降低(均P<0.05)。结论:电针"神门""三阴交"通过抑制交感-肾上腺髓质系统的过度兴奋治疗失眠。
Objective To explore the action mechanism of electroacupuncture(EA)at"Shenmen"(HT 7)and"Sanyinjiao"(SP 6)on insomnia.Methods Thirty SD rats were randomly divided into a blank group,a model group and an EA group,10 cases in each group.The insomnia model was made by immobilization method in the model group and EA group.After model establishment,rats in the blank group and model group were treated with fixation and no treatment was given.Rats in the EA group were treated with EA at"Shenmen"(HT 7)and "Sanyinjiao"(SP 6)for 15 min,once a day for 4days.After treatment,the level of daytime and nighttime activity,open-arm entry percentage and open-arm time percentage of elevated plus-maze test were measured;the content of noradrenaline(NE),dopamine(DA)and epinephrine(EPI)in plasma and NE,DA in thalamus and brainstem were detected by using euzymelinked immunosorbent assay method.Results Compared with the blank group,the daytime activity was increased and nighttime activity was reduced in the model group(bothP〈0.05);the openarm entry percentage and open-arm time percentage of elevated plus-maze test were both reduced in the model group(bothP〈0.05);the contents of NE,DA,EPI in the plasma and NE,DA in thalamus and brainstem were increased in the model group(allP〈0.05).Compared with the model group,the daytime activity was reduced and nighttime activity was increased in the EA group(bothP〈0.05);the open-arm entry percentage and openarm time percentage of elevated plus-maze test were both increased in the EA group(bothP〈0.05);the contents of NE,DA,EPI in the plasma and NE,DA in thalamus and brainstem were reduced in the EA group(allP〈0.05).Conclusion Electroacupuncture at"Shenmen"(HT 7)and"Sanyinjiao"(SP 6)can restrain the over-activity of the sympathetic-adrenal medullary system to treat the insomnia.
