摘要
丝氨酸蛋白酶抑制剂(Serpin)对生物的生长发育、生理功能和病理起到重要调节作用。本试验成功克隆白纹伊蚊丝氨酸抗凝血AL-71基因,序列大小为1 383 bp。经同源性分析,该基因与原序列同源性为88.85%,氨基酸同源性为90.24%。同时,对其分子生物学特性进行预测和建模分析,发现白纹伊蚊的氨基酸序列活性部位和催化结构域表现出较高的相似性。成功构建原核表达载体PET-AL-71,经IPTG诱导,获得分子量大小约为50 KDa的AL-71蛋白,与预期结果一致。为下一步在真核表达系统表达奠定基础。
Serpin plays an important regulatory role in the growth and development of biological,physiological functions and pathology. In this study, we cloned and sequenced the serine anticoagulant AL-71 gene of 1 383 bp. Homology analysis showed that the cloned AL-71 gene had a sequence homology of 84.26% in nucleotides and 82.57% in amino acid. Prokaryotic expression vector PET-AL-71 was successfully constructed to express AL-71 protein.
