淫羊藿苷对Tg2576小鼠学习记忆功能及海马神经细胞再生的影响 被引量:17

摘要 目的观察淫羊藿苷对Tg2576小鼠学习记忆功能及海马齿状回神经细胞再生的作用。方法 Tg2576转基因小鼠及野生型小鼠分为4个组:野生型对照组、野生型+淫羊藿苷组、Tg2576对照组和Tg2576+淫羊藿苷组。给药组灌胃淫羊藿苷60 mg/kg/d,对照组灌胃同体积蒸馏水,每天1次,连续3个月。Morris水迷宫检测动物行为学改变;硫黄素S染色检测海马组织内老年斑;免疫组织化学法检测海马齿状回Brd U+细胞;Western blot检测海马组织淀粉样前体蛋白(APP)和脑源性神经营养因子(BDNF)的蛋白表达。结果淫羊藿苷明显缩短Tg2576小鼠水迷宫逃避潜伏期和搜索距离;明显减少海马组织内的老年斑;增多海马齿状回Brd U+细胞数量;降低海马组织APP高表达和改善BDNF低表达状态。而野生型小鼠给予淫羊藿苷3个月后,上述指标没有显著变化。结论淫羊藿苷改善Tg2576小鼠空间学习记忆功能,并促进海马齿状回神经细胞再生。 Objective To observe the effects of icariin on the learning and memory function and neurogenesis in the dentate gyrus( DG) of hippocampus in Tg2576 mice. Methods Tg2576 mice and wild type mice were divided into four groups: wild type control group,wild type + icariin group,Tg2576 control group,and Tg2576 + icariin group. Icariin- treated mice were administrated with 60 mg / kg / d icariin by gavage. Wild type and Tg2576 mice were given equally volume distilled water,once a day,for 3 months. Morris water maze was used to test the learning and memory function of mice. Senile plaques( SPs) in the hippocampus of Tg2576 mice were observed by thioflavine S staining. The number of Brd U+cells in the DG of hippocampus was detected through immunohistochemical staining. The protein expressions of amyloid precursor protein( APP) and brain derived neurotrophic factor( BDNF) were assayed by western blot. Results Icariin not only significantly reduced the latency and distance of Tg2576 mice in water maze,decreased SPs in the hippocampus of Tg2576 mice,increased the number of Brd U+cells in the DG of hippocampus,but also reduced the expression of APP and elevated the expression of BDNF in the hippocampus of Tg2576 mice. However,the above parameters did not change in icariin- treated wild type mice. Conclusion Icariin improves the function of learning and memory of Tg2576 mice,and promotes neurogenesis in the DG of hippocampus.
