摘要
由灰葡萄孢菌(Botrytis cinerea)引起的灰霉病是一种危害极大的植物病害,每年都会造成作物的严重的损失。但B.cinerea也是贵腐葡萄酒品质的主要重要成因。试验采用生物信息学的方法,对B.cinerea的胞外分泌蛋白进行分析。综合利用信号肽分析软件SignalP 4.1、跨膜结构预测软件TMHMM v2.0和THUMBUP、GPI锚定位点预测软件big-Pi Predictor、亚细胞蛋白定位软件TargetP 1.1等软件,对B.cinerea全基因组中的16448个氨基酸序列进行预测。共发现有628个分泌蛋白,编码这些蛋白的ORF最短333bp,最长4476bp,平均1055bp。367个蛋白质有功能描述,其中包括214个酶类。这些酶主要包括各种糖苷水解酶、酯酶、蛋白酶、多糖聚合酶、氧化还原酶、角质酶等。本文对研究B.cinerea的侵染机制和解释贵腐酒特殊香气的原因有重要意义。
Botrytis cinerea is one of the most devastating plant pathogens,and causes huge losses every year.Wine grapes infected by B.cinereacan produce particularly fine and concentrated sweet wine,named " noble rot".Therefore,the study of B.cinereasecretome posed important meaning to the infection mechanisms and noble rot wine production.In the present study,prediction algorithms SignalP 4.1,TMHMM v2.0,big-PI predictor and TargetP v1.1were applied comprehensively to analysis the total secreted proteins based on the whole genome sequences.Among the 16448 predicted ORFs,628 proteins were secreted and ORF length was 333~4476bp with an average length of 1055 bp.367proteins were annotated with function description,of which 214 proteins were enzymes,and glycoside hydrolases,polysaccharide polymerase,oxidoreductases,esterases,proteases were discovered.These enzymes and proteins provided important information for the infection mechanisms of B.cinereaand useful traits for the production of noble rot wine.
