前列腺癌中NUCB2和caspase-3的表达及其相关性

摘要 目的检测人良性前列腺增生(benign prostatic hyperplasia,BPH)、前列腺上皮内瘤变(prostatic intraepithelial neoplasia,PIN)及前列腺癌(prostate cancer,PC)组织中NUCB2和caspase-3的表达,探讨二者在前列腺癌中表达的意义和相关性。方法应用免疫组化SP法检测在BPH、PIN及PC组织中NUCB2与caspase-3的表达,并分析二者在PC组织中的表达有无相关性。结果 NUCB2在PC组织中的表达显著高于BPH和PIN组织,其表达与年龄无关,与Gleason分级、术前血清PSA有关;<5年生存期病例的NUCB2表达阳性率高于>5年生存期病例(P<0.05)。caspase-3在BPH组织中的表达显著高于PC和PIN组织,其表达与年龄、Gleason分级及血清PSA均无关。<5年生存期病例的caspase-3表达阳性率低于>5年生存期病例,但无统计学差异(P>0.05)。在前列腺癌组织中NUCB2与caspase-3的表达呈负相关(r=-0.567,P<0.001)。结论NUCB2与caspase-3两者间可能存在一种相关性,联合检测二者有助于前列腺癌的进一步研究。NUCB2的检测可能有助于评估前列腺癌的预后。 Objective To explore the expression of NUCB2 and caspase-3 in benign prostatic hyperplasia(BPH),prostatic intraepithelial neoplasia(PIN) and prostate cancer(PC) and its clinical significance.Methods The expression of NU CB2 and caspase-3 in BPH,PIN and PC was observed using the immunohistochemical SP method.The relationship between NUCB2 and caspase-3 in prostate cancer was analyzed.Results The positive expression rate of NUCB2 in PC was significantly higher than those in BPH and PIN.The expression of NUCB2 was associated with Gleason grading and preoperative PSA,but not with the age.NUCB2 expression in patients with survival of less than 5 years was higher than in patients with survival of more than 5 years(P < 0.05).The positive expression rate of caspase-3 in BPH was significantly higher than those in PC and PIN.The expression of caspase-3 in PC was not related with Gleason grading,preoperative PSA and the age.Caspase-3 expression in patients with less than 5 years was lower than in patients with survival of more than 5 years,but there was no significant difference (P > 0.05).Expression of NUCB2 was negatively correlated with caspase-3 (r =-0.567,P < 0.001).Conclusion Expression of NUCB2 and caspase-3 may be related,and their combined detection may be helpful for further study on the prostate cancer.NUCB2 may become a useful target for evaluating the prognosis in PC.
