胃癌中Fascin表达与K-ras突变情况及两者相关性的研究 被引量:3

摘要 目的:观察胃癌组织中Fascin表达和K-ras突变情况,并分析其与临床病理特征的关系,研究Fascin表达和K-ras突变之间的相关性,探讨两者在胃癌浸润转移过程中的作用。方法:在108例胃癌组织中采用免疫组织化学法检测Fascin表达,采用DNA测序法检测K-ras突变。结果:108例胃癌组织中Fascin表达率为25.00%,K-ras突变率为11.11%;胃癌组织中Fascin阳性表达,发生淋巴结转移组多于未发生淋巴结转移组(P<0.05),发生远处转移组多于未发生远处转移组(P<0.01);K-ras突变,发生远处转移组多于未发生远处转移组(P<0.01);胃癌组织中K-ras突变和Fascin表达之间存在相关性,Fascin阳性比Fascin阴性的胃癌组织更倾向于发生K-ras突变(rp=0.378,P<0.01)。结论:在胃癌组织中K-ras基因突变和Fascin蛋白阳性表达存在相关性,且与肿瘤的转移有关。 Objective: To observe mutation of K-ras and expression of Fascin in gastric tissues and their relationship with clinical pathological features, then to analyze the correlation between mutation of K-ras and expression of Fascin. Methods: In 108 cases of gastric cancer tissues K-ras mutation was detected by DNA sequencing,Fascin expression was detected by method. Results: In gastric tissues the mutation rate of K-ras was 11.11%, the expression rate of Fascin was 25.00 %; The mutation rate of K-ras in distant metastasis group was higher than without distant metastasis group (P〈0.01); The expression rate of Fascin in lymph node metastasis group was higher than without lymph node metastasis group (P 〈0.05), and the expression rate of Fascin in distant metastasis group was higher than without distant metastasis group (P〈0.01); There was a correlation between the mutation of K-ras and the expression of Fascin in gastric tissues, Fascin positive gastric cancer tissue are more inclined to K-ras mutation than Fascin negative gastric cancer tissue (rp = 0.378, P〈0.01). Conclusions: In gastric tissues there is a correlation between the mutation of K-ras and the expression of Fascin. Both of them play an important role in the process of invasion and metastasis.
