摘要
目的探讨和厚朴酚对成年大鼠心肌细胞收缩功能的影响及其机制。方法 Langendorff灌流分离的成年大鼠心肌细胞,Ion Optix光谱荧光同时检测心肌细胞收缩与钙瞬变以及和厚朴酚的作用。并通过改变细胞内钙观察心肌细胞收缩的钙敏感性及和厚朴酚的作用。结果和厚朴酚能降低心肌细胞收缩过程中内钙的水平,并抑制心肌细胞收缩,对心肌细胞收缩的钙敏感性不影响。结论和厚朴酚通过降低心肌细胞内自由钙离子而抑制心肌细胞的收缩功能,不影响心肌细胞收缩的钙敏感性。
Objective To explore the effect and mechanism of honokiol on contractile function and calcium transient of adult rat myocardial cells. Methods Adult rat cardiomyocytes were isolated with Langendorff perfuse system. Myocardial cell contraction and calcium transient were detected with Ion Optix fluorescence spectrum detect system. Series of concentration of Ca Cl2 incubation were used to detect the calcium sensitivity of cardiomyocyte contraction. Results Honokiol reduced myocardial cell contraction and intracellular calcium levels,but the calcium sensitivity of myocyte contraction was not affected. Conclusion Honokiol inhibits the cardiomyocyte contractile function as a result of reduction of intracellular free calcium ions. The effect does not involve the calcium sensitivity of cardiomyocyte contraction.
