摘要
目的检测内源性硫氧还蛋白-1(thioredoxin-1,Trx-1)在急性坏死性胰腺炎(acute necrotizing pancreatitis,ANP)大鼠模型血液、胰腺和肾组织的表达;并观察中药清胰Ⅱ号颗粒剂对ANP胰腺和肾脏的保护作用以及对肾脏组织Trx-1表达的影响。方法将96只SD大鼠随机分为正常组(A组,n=24)、假手术组(B组,n=24)、ANP组(C组,n=24)及治疗组(D组,n=24)。在制模后1小时治疗组给予250g/l清胰Ⅱ号颗粒剂灌胃(10ml/kg),6h1次,共3次,正常组、假手术组、ANP组三组给予等量生理盐水灌胃。于制模后6h、12h、24h测定各组血清中Trx-1的水平;制模后24小时处死大鼠,应用免疫组织化方法对胰腺以及肾脏中Trx-1的表达情况进行测定。结果 24小时内正常组、假手术组血清中Trx-1的表达较稳定,ANP组呈现一个持续升高的趋势,而治疗组Trx-1的表达则呈现一个先上升后下降的趋势,12h治疗组Trx-1的表达较其它三组明显升高(P<0.05),24hANP组血清中Trx-1的表达较假手术组明显升高(P<0.05),而24h治疗组血清中Trx-1的表达又较ANP组降低(P<0.05),但仍高于正常组和假手术组(P<0.05);病理结果显示ANP组胰腺及肾脏组织损伤明显,Trx-1的表达呈阳性,较正常组、假手术组、治疗组比较有统计学意义(P<0.05)。结论 Trx-1在ANP模型大鼠肾损伤中可能具有重要作用,中药清胰Ⅱ号颗粒可提前促进胰腺、肾脏组织Trx-1的表达,减轻ANP时胰腺和肾脏损害,对其有保护作用。
Objective To investigate the expression of thioredoxin-1 (Trx-1) in the serum,pancreas and kidneys of rats with sodium taurocholate induced acute necrotizing pancreatitis(ANP) and assess the effects of Qingyi Ⅱ on Trx-1 expression. Meth- ods Ninty-six healthy SD rats were randomly divided into four groups: the normal group(group A,n=24) ,the sham-operated group ( group B, n= 24 ), ANP model group ( group C, n= 24 ) and the treated group ( group D, n = 24 ). Qingyi II was adminis tered, beginning from 1 hour after modeling for three times ( every 6 hours) per day via intragastric in fusion to group D in dose of 10 ml/kg( 250 g/l). While to the other three groups,equal volume of saline was infused instead: All animals were sacri- ficed after modeling. The Trx-1 expression of the serum was determinated on 6 h, 12 h,24 h after modeling. The Trx-1 expres sion of pancreas and kidneys tissues were determinated by immunohis to chemical methods. Results The expression of serum Trx-1 were higher in group D than that in other three groups (P〈0.05) at the time of 12 hours and higher in group C than that in group A and group B( P〈0.05 ) at the time of 24 hours,group C had serious pathologic damage in pancreas and kidneys (P 〈Q0.05 ). Conclusion Qingyi Ⅱ rises the concentration of Trxl in serum, pancreas and kidneys tissue in advance and alleviate the pathologic damage of the pancreas and kidneys in rats with and complicated with kidneys injury, which suggests that Qingyi Ⅱ may play an important role in treating ANP.
