摘要
目的:考察“一测多评”法同步测定黄柏配方颗粒及黄柏饮片中5种相同成分含量的可行性和准确性。方法:利用HPLC法,以小檗碱为内标物,计算与黄柏碱、药根碱、巴马汀和黄柏酮的相对校正因子,通过相对校正因子计算黄柏配方颗粒4种成分的含量并在黄柏饮片中进行验证,比较“一测多评”法测定值与外标法实测定值的相似度。结果:在一定线性范围内,黄柏碱、药根碱、巴马汀和黄柏酮与内标物小檗碱的相对校正因子分别为0.2350,0.8907,0.7398和0.7698,且在不同实验条件下重复性良好。
OBJECTIVE To study feasibility and accuracy of quantitative analysis of multi-components by single marker (QAMS) for simultaneous determination of contents of five components in Phellodendri Chinensis Cortex formula granules and cork pieces. METHODS HPLC was used, with berberine as internal standard, relative correction factors (RCF) of phellodendrine, jatrorrhizine, pamatin and obakunone were calculated. According to RCF, contents of four components were simultaneously measured in PheUodendri Chinensis Cortex formula granules and cork pieces. Similarity was determined between measured values of QAMS method and real values of external standard method. RESULTS Within linear ranges, RCF values of phellodendrine, jatrorrhizine, pamatin and obakunone were 0. 235 0, 0. 890 7, 0. 739 8 and 0. 769 8, respectively. RCF had a good reproducibility in various instruments and chromatographic columns (RSD= 0. 18% - 0. 63 %). Results of QAMS method were validated by comparing with that of external standard method, and no obvious significant differenCe was found. CONCLUSION QAMS method can be used for quantitative analysis and quality evaluation of Phellodendri Chinensis Cortex formula granules and cork pieces.
