摘要
利用高分离度快速液相色谱与四极杆-飞行时间质谱(RRLC-Q-TOF MS)联用技术,对酒制人参和醋制人参的人参皂苷类成分进行比较研究。样品经不同方法炮制,离心过滤后,采用Agilent ZORBAX SB-C18色谱柱,以0.1%甲酸和乙腈作为流动相进行梯度洗脱,采用高分辨质谱进行检测,利用Masshunter Qualitative Analysis软件与人工相结合进行分析。鉴定了46种人参皂苷,比较了酒制人参和醋制人参中人参皂苷含量差异,并检测到稀有人参皂苷。同时总结了炮制品中人参皂苷的转化途径。对人参置于酒或醋中有效成分的浸出量进行比较。本方法对于人参炮制品的成分研究具有借鉴意义,并且对民间使用的人参泡酒进行了科学解释,为人参炮制品在临床合理使用提供指导。
Rapid resolution liquid chromatography coupled with quadrupole-time-of-flight tandem mass spec- trometry ( RRLC-Q-TOF MS/MS) was used for the comparative analysis of ginsenosides composition in white ginseng, vinegar processed ginseng, distilled spirit processed ginseng and millet wine processed ginseng. Under the optimal chromatographic conditions including ZORBAX SB-C18 column (100 mm×2. 1 mm, 3.5μm) at 35℃, 5μL of injection volume and water-acetonitrile gradient elution as mobile phase with a flow rate of 0.3 mL/min, 46 kinds of ginsenosides were identified by the comparison of the retention time of the standard compound and the accurate mass obtained from RRLC-Q-TOF MS/MS. The rare ginsenosides were detected by the comparison of white ginseng with Processed-ginseng, such as ginsenoside Rh2, 172, Rg4, Rgs, Rk3 etc. The method has reference significance for the research of compounds in the processed Chinese-herbs.
