摘要
目的观察中药膏方联合特异性免疫治疗(SIT)对小鼠支气管哮喘的治疗作用。方法将BALB/c小鼠按随机数字表法随机分为空白对照组、哮喘模型组、SIT组、SIT联合中药膏方组,采用卵清蛋白(OVA)腹腔注射及雾化激发构建支气管哮喘小鼠模型,给予SIT及中药膏方灌胃治疗,进行肺泡灌洗液细胞计数和分类,用ELISA法测定血清IgE、肺泡灌洗液IL-4、IFN-γ含量,用HE染色法检测小鼠肺组织病理学改变。结果 SIT、中药膏方联合SIT治疗组小鼠肺组织炎症细胞浸润程度减轻,其中后者作用更显著,血清IgE水平较哮喘组下降(P<0.05),中药膏方组联合SIT组较SIT下降显著;SIT、中药膏方联合SIT组在肺泡灌洗液(BALF)中白细胞总数及嗜酸性粒细胞总数均减少,BALF的IL-4下降、IFN-γ上升(P<0.05),但两者间差异无统计学意义(P>0.05)。结论中药膏方联合特异性免疫治疗可以降低小鼠血清IgE水平,减轻肺部炎症细胞浸润程度。
Objective： To observe the effects of Shengqi Zhuangyang medicinal extract combined with specific immunotherapy on airway inflammation of mouse with bronchial asthma, and to explore its mechanism in treating asthma. Methods： Twenty BALB/c mice were randomly divided into the blank control group,the asthma group, the SIT group and the SIT ＋ medicinal extract group. Asthma was induced by intraperitoneal injection of ovalbu- rain（OVA） and forced inhalation of atomized OVA. The numbers of the total inflammation cells and esoinophils （EOS） in BALF were counted by alveolar wash. IL-4, IFN-γ/in BALF and IgE in blood were analyzed by ELISA. Pathological changes of the lung tissues were observed by HE straining. Results： After the therapy,the lung in- flammation was reduced, and the IgE level in blood was lowered, especially in the SIT ＋ medicinal extract group, and the numbers of total cells and EOS decreased. IFN-γ/level was raised and IL-4 level was lowered in BALF, which had no differences between the SIT group and the SIT ＋medicinal extract group. Conclusion： Shengqi Zhuangyang medicinal extract combined with specific immunotherapy can reduce the inflammation of lung and lower IgE level in blood
