鸡基因工程复合抗病毒制剂冻干保护剂的筛选

摘要 采用不同高压灭菌条件处理的不同比例的脱脂奶粉、蔗糖、海藻糖以及维生素C组成的保护剂配方对鸡基因工程复合抗病毒制剂进行冻干,冻干后通过外观检查、p H值检测、复水性检测及效价测定比较筛选出了理想的冻干保护剂,之后将普通水剂型和冻干型鸡基因工程复合抗病毒制剂分别置于4℃、-20℃、-70℃条件下进行保存期试验,结果表明采用脱脂奶粉+蔗糖、脱脂奶粉+蔗糖+海藻糖+维生素C两种配方对鸡基因工程复合抗病毒制剂的保护效果最好,冻干后效价仍为220;普通水剂和冻干型鸡基因工程复合抗病毒制剂在-70℃保存条件下保存360天内效价均未降低,普通水剂分别于90天、180天后效价开始明显下降,而冻干型鸡基因工程复合抗病毒制剂的效价均无明显降低,稳定性良好。本研究初步筛选出了鸡基因工程抗病毒制剂的冻干配方,显著延长了其保存期,为鸡基因工程抗病毒制剂规模化生产和推广应用奠定了基础。 The chicken genetic engineering composite antiviral preparation was freeze-dried with different cryoprotectant prescriptions made up with skim milk, sucrose, trehalose and Ve by means of different autoclaved sterilization. The ideal cryoprotectant was selected through appearance examination, pH measurement, rehydration test and potency test. The aqueous chicken genetic engineering composite antiviral preparation and lyophilized chicken genetic engineering composite antiviral preparation were placed under 4 ℃, -20 ℃, -70 ℃ for preservation period test. The result suggested that the two cryoprotectant prescriptions (skim milk + sucrose and skim milk + sucrose + trehalose + Vc) could best protect chicken genetic engineering composite antiviral preparation with the antiviral titer of 2^20. The preservation period test showed that the aqueous chicken genetic engineering composite antiviral preparation and the ly- ophilized chicken genetic engineering composite antiviral preparation under -70~C could be preserved for one year with no decline of the titer; while under 4℃ and -20℃, the titers of the aqueous preparation started to decline respectively after 90d and 180d, and the titer of the lyophilized preparation did not decline under -20 ℃ and decreased a little under 4℃ after 180d. The result indicated that the selected cryoprotectants could remarkably extend the preservation period of the chicken genetic engineering composite antiviral preparation. The study selected the cryoprotectant of chicken genetic engineering composite antiviral preparation, successfully prepared lyophilized chicken genetic engineering composite antiviral preparation with good appearance. It also lay the foundation for the large-scale production of chicken genetic engineering composite antiviral preparation and made some contribution to clinic promotion and application.
