摘要
利用RAPD标记对银白杨(♀)×新疆杨(♂)杂交后代及其亲本进行DNA多态性鉴定。结果表明,8对引物共扩增出46条带,平均每对引物扩增出5.75条带,多态性带占扩增总带数的41.3%,扩增片段大小在300~3000bp之间。父母本的特异性条带在子代中出现的频率分别为51.25%和46.88%。杂交后代的带型分为叠加型、缺失型和特异型。父母本之间的遗传相似系数为0.6522,子代8与父本遗传相似系数最高为0.9130,子代6与母本遗传相似系数最高为0.8478。本研究从分子水平鉴定杂交种,为开展分子标记辅助杨树杂交亲本选配和杂种子代表现预测奠定了基础。
RAPD markers were used to identify DNA polymorphism among hybrids from the cross of Populus alba (♀) ×Populus alba var. pyramidalis ( ♂ ) and their parents. The results are as follows : a total of 46 clear and re- producible bands were amplified by 8 RAPD primers, each primer amplified 5.75 bands on average, the frequen- cy of polymorphism was 41.3 %, the bands distributed from 300 - 3 000 base pairs (bp). Special bands of Pop- ulus alba var. pyramidalis and Populus alba Linn. occupied 51.25 % and 46.88 % in offsprings, respective- ly. There were 3 banding patterns in hybrids, namely combined bands, deleted bands, and special bands. The similarity was 0. 652 2 between Populus alba var. pyramidalis and Populus alba Linn. The maximum similarity was 0. 913 0 between offspring ( No. 8 ) and male parent. The maximum similarity was 0. 847 8 between offspring ( No. 6) and female parent. This study gives a preliminary exploration in marker-assisted grouping of parental trees and marker-assisted prediction of hybrid performance in poplar hybridization.
