氨甲蝶呤对映体对肺癌A549细胞的生长抑制作用研究

摘要 目的:研究氨甲喋呤(MTX)对映体对肺癌A549细胞的生长抑制作用。方法:倒置相差显微镜观察加入MTX对映体后细胞形态变化,应用MTT法检测MTX对映体对A549细胞的生长抑制作用,应用流式细胞术分析细胞周期分布及凋亡率的变化。结果:倒置相差显微镜观察加入MTX对映体后细胞形态发生明显变化,MTT法检测表明两种MTX对映体抑制A549细胞的生长呈剂量与时间依赖性,L-(+)-MTX对A549细胞的抑制作用明显强于D-(-)-MTX。流式细胞检测发现两种MTX对映体药物对A549细胞的细胞周期和凋亡具有明显干扰作用。结论:MTX对映体对A549细胞的作用明显具有手性差异,L-(+)-MTX对A549细胞的抑制作用明显强于D-(-)-MTX。 AIM: To study inhibitory effects of methotrexate( MTX) enantiomers in human lung cancer A549 cell lines. METHODS: The cell morphological changes were observed by inverted phase contrast microscopy. Cell growth inhibition was determined by MTT method. The cell cycle phases and apoptosis rates were analyzed by flow cytometry. RESULTS: The cell morphogenesis changes were obvious after joining MTX enantiomers by inverted phase contrast microscopy. The inhibitory effects of MTX enantiomers showed dose and time dependent inhibition of cell growth in human A549 cells. The inhibitory effect of L-( +)-MTX was stronger than that of D-(-)-MTX. There were detectable disruption of cell cycle and apoptosis of A549 cells after MTX enantiomer drug treatments. CONCLUSION: MTX enantiomers produce appreciable chiral selective effect in human A549 cells,the inhibitory effect of L-( +)-MTX is stronger than that of D-(-)-MTX.
