摘要
目的总结近年来国内外具有应用于乳腺癌骨转移潜在靶点能力的药物或制剂的研究进展。方法检索CHKD期刊全文数据库及PubMed检索系统,以乳腺癌、骨转移、靶点治疗和新型药物为关键词,检索主要分布在2009-01-2014-12的乳腺癌骨转移机制及靶点药物研究的文献。纳入标准:1)骨重塑和乳腺癌骨转移的影响因素及相互关系的研究;2)乳腺癌骨转移的潜在治疗靶点;3)乳腺癌骨转移的靶向药物研究;4)中医药方面的相关研究。根据纳入标准,选取43篇文献纳入分析。结果骨是乳腺癌最常见的远处转移部位。乳腺癌骨转移是包括肿瘤细胞和骨骼微环境等多种因素共同作用的结果。近年来涌现的一些新型药物和制剂,如mTOR抑制剂依维莫司、SRC抑制剂塞卡替尼和达沙替尼、组织蛋白酶K抑制剂Odanacatib、抗Sclerostin抗体Romosozumab和中药等,在体外实验和临床研究中表现出了抑制肿瘤骨转移的潜力。结论对乳腺癌骨转移机制的深入研究有助于发现新的药物靶点,并将促成更多药物的开发以用于骨转移的预防和治疗。
OBJECTIVE To review advances both home and abroad in targeted drugs or agents which could be ap- plied in the treatment of breast cancer bone metastases in recent years. METHODS Literature on mechanisms of hone metastases of breast cancer and the research on related targeted drugs were retrieved from PubMed and CHKD database from 2009.1 to 2014.12,with ＂breast cancer,bone metastases, targeted therapy and new drugs＂ as keywords. Retrieval criterion： research on the influencing factors of bone remodeling and bone metastases of breast cancer and their relation- ships~ potential therapeutic targets for bone metastases of breast cancer~ research on potential targeted drugs for bone me- tastases of breast cancer~ related research on traditional Chinese herbal medicine. According to the inclusion criteria, 43 articles were selected for further reviewing. RESULTS Bone is the most common distant metastasis site in breast cancer and bone metastasis results from joint actions of many factors such as tumor cells and bone microenvironment. Some new drugs and agents with the potential of inhibiting bone metastases emerged in recent years, including mTOR-in- hibitors （everolimus）, SRC kinase inhibitors（saracatinib and dasatinib）, Cathepsin K inhibitors （odanacatib）, anti- sclerostin antibody （romosozumab）, herbal medicine and so on. CONCLUSION The increasing understanding of the mechanisms of bone metastasis in breast cancer will help to seek for new therapeutic targets and will contribute to the de- velopment of more drugs for the prevention and treatment of bone metastases.
