摘要
采用超高压技术提取瓜蒌多糖,以瓜蒌多糖得率为指标,考察提取溶剂、超高压压力、保压时间和料液比(g/m L)对瓜蒌多糖得率的影响。正交实验结果显示,瓜蒌多糖最佳提取工艺条件为:以水为提取溶剂,提取压力100 MPa,保压时间3 min,料液比1:40(g/m L),瓜蒌多糖的提取率达19.11%。同加热回流提取和超声波辅助提取法相比,超高压提取得率明显高于超声提取,略低于加热回流提取,但其提取时间分别为加热回流提取和超声波辅助提取的1/10和1/20,提取时间短,是提取瓜蒌多糖的适宜方法。同时,采用黏度计对超高压提取的瓜蒌多糖黏度性质进行了表征,研究了瓜蒌多糖溶液的浓度、温度、p H值及金属离子对其黏度的影响,结果表明,超高压提取的瓜蒌多糖溶液黏度很低,浓度、温度、p H值及金属离子对其黏度都有一定的影响。
Extraction of poiysaccharide from Trichosanthes kiriiowii Maxim.Using ultrahigh pressure extraction(UPE) was studied.The experiment was optimized with the orthogonal test design.The optimum extraction conditions were 100 MPa of extraction pressure,3 min of extraction time and 1:40(g/mL) solid/liquid ratio in water solvent.Under the optimized conditions,the yield of poiysaccharide reached 19.11% .Compared with the heat reflux extraction(HRE) and the ultrasonic-assisted extraction(UAE),UPE has higher extraction yield than UAE.Besides,the extraction time of UPE is only 3 min which is1/10 and 1/20 of HRE and UAE,respectively.It can be considered as an alternative extraction technique for fast extraction of poiysaccharide from Trichosanthes kiriiowii Maxim.The effects of poiysaccharide concentration,temperature,pH value and metallic ions on viscosity of Trichosanthes kiriiowii Maxim,poiysaccharide were examined in this study.The results show that the viscosity is very low and these factors could affect its viscosity to differ extent.
