摘要
目的:观察潜阳育阴颗粒(Qianyang Yuyin granules,QYG)对正常麻醉犬肾脏微循环影响,并初步研究其作用机制。方法:应用Power Lab系统记录十二指肠给予潜阳育阴颗粒前和给药后15~180 min不同时段比格犬肾脏微循环变化;犬分为正常组,QYG高、中、低剂量组(12.30,6.15,3.08 g·kg-1),卡托普利组(30 mg·kg-1)和珍菊降压片组(0.14 g·kg-1),于给药前及给药后15,30,60,90,120,180 min通过股动脉取血,ELISA法检测血清中血管紧张素Ⅱ(AngⅡ),内皮型一氧化氮合酶(e NOS)含量,化学法检测一氧化氮(NO)含量;给药3 h后取肾脏组织,ELISA法检测肾脏AngⅡ含量。结果:潜阳育阴颗粒12.30 g·kg-1剂量组给药后60~180 min正常麻醉犬的肾动脉微循环血流量明显上升(P〈0.05),给药180 min后肾脏组织AngⅡ含量明显下降(P〈0.05),给药90,120,180 min后,血清中AngⅡ明显下降(P〈0.05),e NOS和NO含量明显上升(P〈0.05);潜阳育阴颗粒6.15 g·kg-1剂量组给药后90~120 min内正常麻醉犬的肾动脉微循环血流量明显上升(P〈0.05),给药90,120 min后,血清中AngⅡ明显下降(P〈0.05),e NOS和NO含量明显上升(P〈0.05);潜阳育阴颗粒低剂量组给药后180min内正常麻醉犬各项指标变化不明显。结论:潜阳育阴颗粒可改善肾脏微循环,其作用机制可能与降低循环及肾脏组织AngⅡ的含量,增加循环中e NOS及NO含量有关。
Objective: To study the effects of Qianyang Yuyin granules( QYG) on renal microcirculation in normal dogs and to investigate the mechanism. Method: The renal microcirculation was determined by Power Lab system before giving QYG( 3. 08,6. 15,12. 30 g·kg-1) and after 15,30,60,90,120,180 min respectively. Result: Compared with the normal group,QYG( 12. 30 g·kg-1) could increase the renal microcirculation( P 〈 0. 05,60-180 min),the contents of nitric oxide synthase( e NOS)( P 〈 0. 05) and nitric oxide( NO) in the serum( P 〈 0. 05,90-180 min) and reduce the contents of angiotensinⅡ( AngⅡ) in the serum( P 〈 0. 05,90-180 min) and kidney( P 〈 0. 05,180 min); QYG( 6. 15 g·kg-1) could increase the renal microcirculation( P 〈 0. 05,90-120 min),the contents of e NOS and NO in the serum( P 〈 0. 05,90-120 min) and reduce the contents of Ang Ⅱ in the serum( P 〈 0. 05,90-120 min). The effects of QYG( 3. 08g·kg-1) were not obvious. Conclusion: QYG could improve renal microcirculation,its mechanism may be associated with the decreasing circulation and kidney tissue AngⅡ content,increasing the circulation of e NOS and NO contents.
