摘要 目的探讨白细胞介素18(IL-18)、肿瘤坏死因子α(TNF-α)对3T3-L1脂肪细胞、RAW264.7巨噬细胞的白细胞介素18受体β(IL-18Rβ)表达的影响。方法采用反转录PCR和Western blot法检测不同浓度IL-18、TNF-α处理后3T3-L1脂肪细胞、RAW264.7巨噬细胞的IL-18RβmRNA及蛋白的表达。结果 10 ng/m L、100 ng/m L TNF-α显著增加3T3-L1脂肪细胞IL-18Rβ的mRNA及蛋白表达,10 ng/m L、100 ng/m L IL-18显著增加RAW264.7巨噬细胞IL-18Rβ的mRNA及蛋白表达。结论IL-18、TNF-α可能通过促进巨噬细胞、脂肪细胞IL-18R的表达参与糖尿病和肥胖的脂肪组织慢性炎症过程。 Objective To explore the effects of interleukin 18(IL-18) and tumor necrosis factor alpha(TNF-α) on the expression of IL-18 receptor beta( IL-18Rβ) in 3T3-L1 adipocytes and RAW264. 7 macrophages. Methods The level of IL-18Rβ was determined by reverse transcription PCR and Western blotting in both 3T3-L1 adipocytes and RAW264. 7 macrophages after processed by different concentrations of IL-18 and TNF-α. Results The level of IL-18Rβ mRNA and protein significantly increased in 3T3-L1 adipocytes after treated with 10 ng / m L,100 ng / m L TNF-α. The expression of IL-18Rβ was also raised in RAW264. 7 macrophages after treated with 10 ng / m L,100 ng / m L IL-18. Conclusion IL-18Rβ could be up-regulated by IL-18 or TNF-α in 3T3-L1 adipocytes and RAW264. 7 macrophages. IL-18 may be an important factor involved in the inflammation of adipose tissues in diabetes and obesity.
