摘要
目的研究IL-23在体内外对肝细胞肝癌发展的影响。方法采用慢病毒体系将构建好的IL-23质粒转入Hepa1-6肝癌细胞株中,流式分选得到过表达IL-23的Hepa1-6稳转细胞,ELISA确定所得到的稳转细胞能够表达高水平的IL-23。采用MTT方法检测IL-23对Hepa1-6细胞增殖的影响,流式细胞染色法检测IL-23对Hepa1-6细胞周期的影响,通过原位种植Hepa1-6细胞建立肝癌模型,检测IL-23体内在肝细胞肝癌发展中的作用。结果 IL-23能够明显促进肝癌细胞的增殖,然而对肝癌细胞的凋亡却没有影响,IL-23在体外促进肝癌细胞的迁移水平,并且在小鼠肝癌模型中,IL-23在体内能够促进肝癌的发展。结论IL-23在体内体外能够直接促进肝细胞肝癌的发展。
To observe the impact of IL-23 on the development of hepatocellular carcinoma(HCC) cells in vitro, stably transfected Hepa1-6-IL-23 cells that can stably secret IL-23 were generated and confirmed by detecting the expression of IL-23 with ELISA assay. We applied MTT assay to detect cell proliferation, flow cytometry to detect the expression of Ki67 and cell cycle. Data showed that IL-23 significantly promoted the proliferation of HCC cells while did not affect the apoptosis. The percent of cells in G2/S phases was also increased by IL-23; IL-23 also increased the mobility of HCC cells in vitro. Moreover, IL-23 expression facilitated the HCC growth in vivo. Our study provides evidence that IL-23 could directly promote HCC development through increasing HCC cell proliferation.
