摘要
为探讨乳房链球菌(Streptococcus uberis)感染乳腺上皮细胞后产生的炎症变化及牛磺酸对炎性变化的缓解作用,以小鼠乳腺上皮细胞系Ep H4-Ev和乳房链球菌0140J为研究对象,检测二者共孵育后细胞产生的炎性变化;在此基础上探究牛磺酸对乳房链球菌对Ep H4-Ev黏附及其造成的炎性损伤的影响。结果表明:牛磺酸处理对乳房链球菌的黏附有一定的抑制作用,但不同浓度牛磺酸处理差异不显著(P>0.05)。乳房链球菌感染Ep H4-Ev 2 h后,细胞培养上清液中肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)mRNA表达水平及N-乙酰-β-D-氨基葡萄糖苷酶(NAGase)的活性显著提高(P<0.01);4 h后乳铁蛋白(LF)mRNA表达水平显著上升(P<0.01)。牛磺酸处理小鼠乳腺上皮细胞3 h,可显著降低TNF-α、IL-1β和IL-6的表达水平和NAGase活性(P<0.01)。研究表明:牛磺酸可以减弱因乳房链球菌感染而引起的小鼠乳腺上皮细胞的炎性变化,该作用可能与其降低乳房链球菌对Ep H4-Ev的黏附有关。
The aim of this study is to investigate the cell inflammation changes after mouse mammary epithelial cell line Ep H4- Ev was infected with Streptococcus uberis and the effect of taurine attenuation on inflammation changes. The cell inflammatory changes were detected after Ep H4- Ev incubation with S. uberis,and then the protective effect of taurine on adhesion of S. uberis and cell inflammatory injury was investigated. The results showed that S. uberis could adhere to Ep H4- Ev cells,and taurine treatment could reduce the adhesion,but the different concentrations of taurine had no statistical significance( P〉0. 05). At 2h after inoculation of Ep H4- Ev with S. uberis,the expression level of TNF- α,IL- 1β and IL- 6 mRNA and N- acetyl- β- D- glucosaminidase( NAGase) activity in cell culture supernatants were increased significantly( P〈0. 01); at 4h after infection,the expression level of lactoferrin mRNA was increased significantly( P〈0. 01). The taurine pre-treated mouse mammary epithelial cells significantly reduced the expression levels of TNF- α,IL- 1β and IL-6,and decreased the activity of NAGase( P〈0. 01). It suggested that taurine could attenuate the inflammation changes of mouse mammary epithelial cells induced by S. uberis,and this attenuation may be related to the reduction of the adhesion of S. uberis to Ep H4- Ev.
