摘要
为得到一种高效的适用于两栖动物的基因提取方法,以臭蛙属6个物种的肌肉组织为材料,通过充分破碎组织、频繁更换双蒸水、加快消化速度和低温沉淀加速DNA析出等方法,对常规的DNA提取方法(酚-氯仿抽提法)加以改进.改进后的DNA提取过程耗时缩短至12 h,提取的基因组DNA适合长片段PCR扩增,经琼脂糖凝胶电泳检测,基因组DNA和12S、16S rDNA片段均得到清晰、明亮、整齐的条带,证实该方法在提取基因组DNA时,具有快速、准确、可靠的特点.
To get a highly efficient method of DNA extraction for amphibian,the muscle tissues from six species of genus Odorrana were adopted as experimental materials. The traditional method of phenol-chloroform extraction was improved by some procedures,including fully crushing organization,changing dd H2 O frequently,accelerating the digestion and cryoprecipitation. In general,the improved method took only about 12 h and the extracted DNA can be used to amplify the long genes. By using agarose gel electrophoresis detection,the 12 S and 16 S rDNA fragments of genomic DNA were clear,bright and neat,which confirmed the method in extracting genome DNA was fast,accurate and reliable.
