摘要
目的观察ApoE-/-小鼠动脉粥样硬化(AS)时的形态学、血清学、细胞因子、血管细胞黏附分子-1(VCAM-1)的表达与单核细胞的聚集情况以及川芎对AS的影响,并对其作用机制进行初步探讨。方法将30只ApoE-/-小鼠分为阳性对照组(n=10)、阴性对照组(n=10)及药物干预组(n=10),阴性对照组饲以正常饮食,其余两组饲以高脂饮食,制造AS模型。16周后处死动物,取血清检测血脂水平。取主动脉行HE、油红染色,并针对动脉壁内的CD68、抗平滑肌抗体α(SMA-α)、肿瘤坏死因子α(TNF-α)、VCAM-1行免疫组化染色。采用SPSS 11.0软件对数据进行分析。结果川芎组ApoE-/-小鼠动脉斑块-内膜面积及斑块-管腔横截面积比为0.362±0.038、0.094±0.043,小于对照组的0.533±0.030、0.238±0.053(t值分别为5.75、4.36,均P<0.01)。川芎组高脂饮食ApoE-/-小鼠的血清高密度脂蛋白水平为(0.439±0.071)mmol/L,明显高于对照组的(0.306±0.075)mmol/L(t=8.47,P<0.05)。川芎组ApoE-/-小鼠动脉壁中TNF-α及VCAM-1的表达和内膜下单核巨噬细胞表达比例为0.161±0.046、0.343±0.053、0.381±0.085,明显少于对照组的0.247±0.080、0.474±0.078、0.482±0.047(t值分别为3.58、4.35、3.54,均P<0.05)。而两组间血清总胆固醇、甘油三酯及低密度脂蛋白无明显差异。结论川芎可明显抑制高脂饮食ApoE-/-小鼠AS的发展。川芎可显著提高高密度脂蛋白水平,这可能是川芎抗AS的机制之一。川芎的抗AS作用可能与抑制炎症反应、抑制动脉壁VCAM-1的表达,从而减少单核-巨噬细胞在动脉壁的黏附与聚集有关。
Objcetive To observe the change of morphology ,serum lipoprotein,cytokines,vascular cell adhesion molecule-1 ( VCAM-1 ) expression and monocyte aggregation on ApoE -/-mice with atherosclerosis ( AS) .Verify the inhibition and mechanism of Chuanxiong to AS .Mtehods Thirty ApoE-/-mice were randomly divided into normal diet group (n=10),high-cholesterol diet(n=10) and Chuanxiong group(n=10).After 16 weeks,the animals were killed.The level of serum lipoprotein was detected .Aortas tree were prepared for HE staining,oil red staining,and immunohistochemical staining for CD68,anti-smooth muscle antibodyα(SMA-α),tumor necrosis factorα(TNF-α),VCAM-1.Then graphic analysis software was used to make semi-quantitative analysis .The data were analyzed by SPSS 11.0 software.Results The intimal area and plaque and lumen cross-sectional area ratio of Chuanxiong group ApoE -/-mice atherosclerosis plaque , was 0.362 ±0.038 , 0.094 ±0.043 , less than that of the control group 0.533 ± 0.030,0.238 ±0.053(t=5.75,P〈0.01; t=4.36,P〈0.01).The serum high density lipoprotein (HDL) level of Chuanxiong group and high fat diet ApoE -/-mice was(0.439 ±0.071 ) mmol/L,significantly higher than that of the control group [(0.306 ±0.075)mmol/L](t=8.47,P〈0.05).The expression of TNF-α、VCAM-1 and macrophages in atherosclerosis plaque of Chuanxiong group was 0.161 ±0.046,0.343 ±0.053 , 0.381 ±0.085 ,significantly less than the control group 0.247 ±0.080 ,0.474 ±0.078 ,0.482 ±0.047 ( t=3.58,P〈0.05;t=4.35,P〈0.05;t=3.54,P〈0.05).There were no significant differences of in serum TC,CHO and LDL between the two groups .Conclusion Chuanxiong can inhibit the development of AS on high-cholesterol diet ApoE -/-mice.Chuanxiong can significantly increase the level of HDL ,which may be one mechanism of the inhibition of Chuanxiong .Inhibition of inflammation , depression of arterial wall expression of VCAM-1,thereby,reducing adhesion and aggregation of mono-macrophages to the arterial w
