摘要
为了研究瘤胃厌氧真菌的纤维降解特性,试验采用体外亨氏滚管培养技术,从绵羊瘤胃液中分离得到3株(K、R、N)具有纤维降解能力的厌氧真菌,运用传统形态学并结合ITS序列同源性分析对其进行鉴定。结果表明:K为Neocallimastix属菌,R为Piromyces属菌,N为Cyllamyces属菌。同一菌株在36小时时降解率均极显著低于72小时(P<0.01),在36小时时菌株K、R、N三者间差异不显著(P>0.05),72小时时菌株K的降解率显著高于菌株R和菌株N(P<0.05)。
To study fibre degradation characteristics of rumen anaerobic fungi,three strains( K,R and N) of anaerobic fungi with the ability to degrade the fiber were isolated and obtained from rumen fluid of Inner Mongolia sheep using the Hungate roll tube method. The three strains were identified using traditional morphology combined with the homology analysis of internal transcribed space( ITS) sequence. The results showed that the three strains including K,R and N strains were identified as Neocallimastix spp,Piromyces spp,and Cyllamyces spp,respectively. The degradation rate at 36 h was highly significantly lower( P〈0. 01) than that at 72 h in the same strain. There were no significant differences( P〉0. 05) in the degradation rates at 36 h among the K,R and N strains,but the degradation rate of strain K was significantly higher( P〈0. 05) than that of the R and N strains.
