摘要
目的探讨虫草素对腹腔注射环磷酰胺所致的小鼠骨髓抑制的治疗作用。方法将30只小鼠随机分为空白对照组(A组)、环磷酰胺骨髓抑制组(B组)和虫草素干预骨髓抑制组(C组),每组10只。以5 m L/kg剂量向A组小鼠腹腔注射生理盐水12 d;以50 mg/kg剂量向B组小鼠腹腔注射环磷酰胺7 d后,腹腔注射生理盐水5 d;以50 mg/kg剂量向C组小鼠腹腔注射环磷酰胺7 d后,以25 mg/kg剂量腹腔注射虫草素5 d。对各组小鼠的外周血细胞以及股骨中的骨髓有核细胞进行计数,采用Western blotting检测白细胞中的着丝粒蛋白Cenp B的蛋白表达水平,采用RT-PCR检测Cenp B的m RNA表达水平。结果与A组相比,B组小鼠外周血白细胞、红细胞、血小板,股骨中骨髓有核细胞数量,Cenp B蛋白相对含量及其表达水平均明显降低(P<0.05);与B组相比,C组中除红细胞数量外,其他5项指标均明显增加(P<0.05)。结论虫草素对环磷酰胺所致的骨髓抑制有一定治疗作用。
Objective To investigate the effect of cordycepin on myelosuppression mice caused by intraperitoneal injection of cyclophosphamide. Methods Thirty mice were divided into the control group（ group A）,myelosuppression induced by cyclophosphamide group（ group B） and myelosuppression intervened by cordycepin group（ group C）,10 in each group. Mice in group A were intraperitoneally injected of normal saline for 12 d with the dosage of 5 m L / kg;M ice in group B were injected of normal saline for 5d after injection of cyclophosphamide for 7 d with the dosage of50 mg / kg; M ice in group C were injected of cordycepin for 5 d with the dosage of 25 mg / kg after injection of cyclophosphamide for 7 d with the dosage of 50 mg / kg. The numbers of cells in peripheral blood and nucleated cells in femur of each mouse were counted. The content and the expression level of Cenp B in leukocytes were detected by Western blotting and RT-PCR. Results Compared with those in group A,the numbers of leukocytes,red blood cells,platelets and nucleated cells,and the content and the expression level of Cenp B were significantly decresed（ P〈0. 05）. Compared with those in group B,all the indexes,except for the number of red blood cells,were significantly incresed（ P〈0. 05）. Conclusion Cordycepin has a certain therapeutic function to myelosuppression mice caused by cyclophosphamide.
