摘要
目的研究红花提取物对Ⅰ型、Ⅱ型5α-还原酶的抑制活性。方法基于高效液相色谱(HPLC)法,建立抑制5α-还原酶体外模型,从雌性大鼠肝脏、雄性大鼠附睾组织中提取Ⅰ型、Ⅱ型5α-还原酶,与底物睾酮(T)及供氢体还原型辅酶Ⅱ(NADPH)共同组成了一种微量的酶促反应体系。采用HPLC法测定反应前后T含量,以此判断5α-还原酶的活性。非那雄胺作阳性对照药。结果与正常组比较,红花提取物0.1 g/L剂量组Ⅰ型、Ⅱ型酶活力,T转化率均无显著性差异(P>0.05);0.1,0.5,2.5 g/L剂量组酶活力、T转化率均显著降低(P<0.01)。结论红花提取物对Ⅰ型、Ⅱ型5α-还原酶活性均有一定的抑制作用,可为临床治疗前列腺增生、雄激素脱发等疾病提供参考。
Objective To study the inhibitory effect of Carthamus tinctorius L. extract on type Ⅰ and Ⅱ 5α- reductase activity. Methods The 5α- reductase inhibition in vitro model was established. TypeⅠ and Ⅱ 5α- reductase extracted from the livers of female rats and prostates of male rats,testosterone( T) as a substrate and NADPH as a hydrogen supplier jointly composed a microscale enzymatic reaction system. The concentration of T which measured by HPLC could show the activity of enzyme. The HPLC method was adopted to detect the T content before and after reaction and the 5α- reductase activity was judged by the T content with finasteride as the positive control agent. Results The activity of type Ⅰ and Ⅱ 5α- reductase and the T conversion rate in the Carthamus tinctorius L. extract0. 1 g / L dose group had no statistically significant difference compared with the normal group( P〉0. 05); the activity of 5α- reductase and the T conversion rate in the 0. 1,0. 5,2. 5 g / L dose groups were significantly decreased( P〈0. 01). Conclusion The extract of Carthamus tinctorius L. has certain inhibiting effect on the activity of type Ⅰ and Ⅱ 5α- reductase,which can provide reference for its application for treating prostatic hyperplasia and androgenic alopecia in clinic.
