益气活血方对非破裂型腰椎间盘突出退变髓核细胞ColⅡ、Aggrecan及TIMP-1 mRNA的影响 被引量:11

摘要 目的探讨益气活血方对人体非破裂型腰椎间盘突出退变髓核细胞中Ⅱ型胶原(ColⅡ)、聚集蛋白聚糖(Aggrecan)及金属基质蛋白酶抑制剂-1(TIMP-1)mRNA的表达的影响。方法选择非破裂型腰椎间盘突出症(LDH)的住院患者经手术摘除的腰椎间盘组织,采用组织块法培养原代细胞;以SPF级雄性SD大鼠制备药物血清。将传代第1代的退变髓核细胞分为3组,实验组分别予体积比为5%、10%的益气活血方血清DMEM培养液;对照组加入生理盐水血清DMEM培养液。通过Realtime PCR检测ColⅡ、Aggrecan、TIMP-1mRNA的表达。结果与生理盐水组比较,益气活血方血清可上调非破裂型腰椎间盘突出退变髓核细胞中ColⅡ、Aggrecan mRNA的表达(P<0.05),未检测出TIMP-1mRNA的变化。结论益气活血方可能通过影响非破裂型腰椎间盘突出髓核细胞中ColⅡ、Aggrecan含量,进而发挥防治非破裂型腰椎间盘突出髓核细胞的退变作用。 OBJECTIVE To investigate the effects of serum containing Yiqi Huoxue Recipe, a compound Chinese herbal medicine , on expressions of aggrecan ,TIMP-1 and type Ⅱ collagen mRNAs of the degene rated intervertebral disc cells in pa- tients with unruptured Lumbar intervertebral disc herniation. METHODS Three inpatients diagnosed as unruptured Lumbar intervertebral disc herniation and undergoing lumbar decompressionand fusionsurgery to remove the degenerated disc tissue were enrolled in this study. Primary intervertebral disc cells were isolated using small tissue mass method. Serum containing Yiqi Huoxue Recipe were obtained after SD rats were fed with Yiqi Huoxue Recipe. In the following tests, the first passage cells were cultured with the sera containing drugs and sodiumchloride (NaC1) , respectively. Proliferation of intervertebral disc cells was detected by expressions of type Ⅱ collagen,aggrecan and TIMP-1 mRNAs were detected by realtime polymerase chain reaction. RESULTS Compared with the same concentrations of NaC1 solution, serum containing Yiqi Huoxue Recipe promoted cell proliferation and expression of type Ⅱ collagen and aggrecan mRNA (P〈0.05). but neither promoted nor restrain cell proliferation and expression of TIMP-1 mRNA (P〉0.05). CONCLUSION Yiqi Huoxue Recipe inhibits degeneration of the intervertebral disc cells by regulatingcollagen and proteoglycan expressions.
