摘要
目的通过模拟舌癌局部的酸性环境,加入雷公藤多甙(TWP),检测其对单核/巨噬细胞杀伤肿瘤细胞功能的影响。方法在p H值6.6,6.8酸性环境中,人单核/巨噬细胞THP-1与人舌鳞癌细胞Tca8113共同培养,经不同浓度的TWP干预,MTT法检测Tca8113细胞数量变化,观察此舌癌酸性环境下TWP对单核/巨噬细胞杀伤舌癌细胞作用的影响。结果酸性环境下,经不同浓度TWP干预,THP-1细胞与Tca8113细胞共同培养4小时,THP-1细胞杀伤活性随TWP浓度增加而增大,与浓度成正相关(P<0.05);p H值为6.6,TWP浓度为5×10-1μg/m L时,THP-1细胞杀伤活性随时间延长而增加,与时间成正相关(P<0.05)。结论体外模拟的舌癌酸性环境中,经一定浓度的雷公藤多甙干预,可使单核/巨噬细胞杀伤活性较未干预组增强,与药物浓度及时间成正相关。
Objective To investigate the effects of tripterygium wilfordii polyglycosidium (TWP) on the cytotoxicity function of mononuclcar-maerophagesin vitro acid environment of squamous-cell carcinoma of tongue. Methods MTT assay was used totest the effects on the cytotoxicity of THP-1 monocytes which were interfered by different concentrations of TWP,and co-cuhured with Tca8113 cells in vitro acid environments (pH 6.6 or 6.8). Results MTT assay showed thatinterfered by different concentrations of TWP and co-cultured with Tca8113 cells for 4h in the identical vitro acidenvironment( pH 6.6 or 6.8) ,the cytotoxicity of THP-1 monocytes in- creased as the concentration of TWP increased, and was significantly positively related to the concentration of TWP (P 〈 0. 05). The cy- totoxicity also increased with time ( P 〈 0.05 ) at pH6.6 and TWP of 5 × 10 - 1 p,g/mL,which was significantly positively related to time ( P 〈 0.05 ). Conclusions Cultured in vitro acid environment of squamous-cell carcinoma of tongue, the eytotoxicity function of mono- nuclear-macrophages interfered by TWP of certain concentration increased compared to those with no interference, and presented signifi- cantly positive correlations with TWP concentrations and time.
