摘要
目的观察川椒方对小鼠变态反应性结膜炎P物质(SP)的影响。方法将72只Balb/c小鼠随机分为6组,即正常组,模型组(给予等体积的生理盐水灌胃)、阳性药物组(给予0.1%奥洛他定滴眼液点眼)、川椒方水煎剂低、中、高剂量组(分别给予川椒方5.25,10.5,21.0 g·kg^(-1)灌胃,每日1次,灌胃体积均为10ml·kg^(-1)),每组各12只。用鸡卵清白蛋白免疫小鼠建立变态反应性结膜炎模型。HE染色,行结膜嗜酸性粒细胞计数,ELISA检测小鼠血清中SP的表达水平,免疫组织化学法观察结膜组织中SP的表达。结果与正常组相比,模型组血清中SP表达明显升高(P<0.01),结膜组织中嗜酸性粒细胞数量和SP的表达亦明显升高(P<0.01)。与模型组比较,阳性药物组和川椒方各组血清中SP的表达显著降低(P<0.01);结膜组织中嗜酸性粒细胞数量和SP表达亦明显降低(P<0.01)。结论川椒方治疗变态反应性结膜炎的作用机制可能是通过抑制P物质的表达。
OBJECTIVE To observe the effects of Chuanjiao Decoction (CJD) on substance P (SP) in aller-gic conjunctivitis (AC) with mice. METHODS Seventy-two Balb/c mice were randomly divided into the normal group (12 mice), the model group (12 mice), the 0.1%olopatadine eye drops group (12 mice), the low, medium, and high dose CJD groups (12 mice). After induced allergic conjunctivtis by intraperitoneal injection ovalbumin, mice in the normal group were not gastrogavage, mice in the model group were given 10 mL&#183;kg-1 normal saline by gastrogavage, once a day for 7 days. Mice in the 0.1%olopatadine eye drops group were given 0.1%olopatadine eye drops twice a day for 7 days. Mice in the low, medium, and high dose CJD groups were given CJD 10 mL·kg-1 at the dose of 5.25, 10.5 and 21.0 g·kg-1 by gastrogavage, once a day for 7 days. The concentration of prostaglandin SP in mice serum was measured by ELISA, and expression of SP was detected using immunohistochemisty assay. The num-ber of eosinophils in the mice ’s conjunctiva was count by H&amp;E. RESULTS Compared with the normal group, the expression of SP in mice serum and conjunctiva was significantly increased in the model group (P&lt;0.01), so was the number of eosinophils in the mice’s conjunctiva (P&lt;0.01). Compared with the model group, the expression of SP in mice serum and conjunctiva significantly decreased in the CJD and 0.1%olopatadine eye drops groups (P&lt;0.01);and the number of eosinophils in the mice’s conjunctiva significantly decreased in the model group (P&lt;0.01). There was no significant difference among the CJD groups (P&gt;0.05). CONCLUSIONS The action mechanism of CJD in treating AC might be achieved through inhibited SP.
