摘要
目的:观察眼针运动疗法对脑缺血再灌注(MCAO)模型大鼠大脑缺血半暗带区域脑组织内Ang-1、Tie2表达水平的影响,探讨眼针运动疗法对MCAO模型大鼠大脑缺血半暗带区域内新生血管影响的机制。方法:采用随机数字法,将100只SD大鼠随机分为3组:空白对照组(10只)、假手术组(10只)、模型复制组(80只)。对模型复制组大鼠采用线栓法进行脑缺血再灌注模型复制,再将模型复制成功的大鼠66只(模型复制80只,失败13只,模型评价1只)随机分别为:模型对照组(17只);头针组(17只);眼针组(16只);眼针运动组(16只)。分别进行头针、眼针及眼针运动进行干预治疗。头针组取穴百会、曲鬓,眼针组取穴定位参照人体取穴方法,取肝区、上焦区、下焦区、肾区。眼针运动疗法组眼针干预同时,在跑步机上进行训练。采用酶联免疫吸附(ELISA)法,检测各组大鼠血清中Ang-1含量,采用RT-PCR及western-blot法对各组大鼠脑组织中Ang-1、Tie2基因及蛋白表达水平进行检测。结果:与空白组比较,模型组、眼针组、头针组、眼针运动组大鼠缺血半暗带区域脑组织中Ang-1、Tie2表达水平显著上调(P<0.01);与模型组比较,眼针组和眼针运动组Ang-1、Tie2表达水平显著上调(P<0.05)。与空白组比较,模型组、眼针组、头针组、眼针运动组大鼠缺血半暗带区域脑组织中Ang-1、Tie2 mRNA表达水平显著上调(P<0.01);与模型组比较,眼针组和眼针运动组Ang-1、Tie2 mRNA表达水平显著上调(P<0.05)。结论:眼针运动疗法在一定程度上能促进血清中Ang-1含量,增加Ang-1及其mRNA表达增加,与受体Tie-2结合,启动Ang/Tie-2信号传导系统,在时间和数量上调节缺血半暗带新生血管和侧支循环的形成,改善缺血半影区的脑血流,从而抑制神经元凋亡,恢复脑血流量,促进脑缺血后神经功能的恢复。
Objective: We observe eye acupuncture combined exercise therapy on cerebral ischemia reperfusion (MCAO) model rat brain ischemia half dark area in the brain tissue,Ang -1 and Tie2 expression levels,the influence of eye acupuncture combined exercise therapy on MCAO model rat brain ischemia half dark mechanism with new blood ves- sels in the area. Methods : We used the method of digital meter to divide 100 SD rats into three groups : blank control group (10) and control group (10), model replication group (80). Copy the model group rats by using line switch on cerebral ischemia reperfusion model replication,and then to copy a successful model rats 66 (model replication 80,13 failure, model evaluation 1 only) were randomly divided into the model control group (17), head needle group (17), eye acu- puncture group ( 16), eye acupuncture combined exercise therapy group (16). Head acupuncture needle group took "Baihui" (DU20) and "Qubin" ( GB7 ). Eye acupuncture group found positioning reference to human, taking liver area, the upper energizer area and lower area and kidney area. Eye acupuncture combined exercise therapy group at the same time used the treadmill training. Using ELISA method, test each Ang - 1 content in serum of rats, and by RT - PCR and western - blot method for each group Tie2 Ang - 1 gene and protein expression level for testing. Results : Compared with blank group,the model group, the eye acupuncture group and eye acupuncture combined exercise therapy group's Tie2 and Ang - 1 expressions in brain half dark area increased significantly ( P 〈 0.01 ). Compared with model group, eye acu- puncture group and eye acupuncture combined exercise therapy group's Ang- 1 and Tie2 expressions increased signifi- cantly (P 〈 0.05). Compared with the blank group, the model group, the eye acupuncture group and eye acupuncture combined exercise therapy group's Ang - 1 and Tie2 mRNA expressions increased significantly ( P 〈 0.01 ).
