摘要
利用巢式PCR(nestPCR)克隆大鼠CHOP基因.该基因ORF区全长507bp,编码168个氨基酸,推测其分子量为19.03kDa,等电点4.59.大鼠CHOP蛋白在第95~160位氨基酸残基位点存在一个“碱性亮氨酸拉链”结构域.蛋白进化分析显示:大鼠CHOP蛋白与啮齿类该蛋白具有较近的亲缘关系.实时荧光定量PCR结果显示:大鼠早期颅脑损伤中,CHOP基因表达呈逐步上升趋势,颅脑损伤24h表达量达到顶峰,为对照组的11.47倍.
A full length cDNA of rat CHOP gene was isolated by nest PCR method. The ORF of CHOP was 507 bp, encoding 168 amino acid residues. The deduced molecular weight and PI were 19. 03 kDa and 4. 59, respectively. A "basic region leucin zipper" domain was found in 95-160 amino acid residues of rat CHOP. The protein phyletic evolution analysis indicated that rat CHOP protein is a close relative of rodents CHOP protein. The realtime qPCR result indicated that the expression level of CHOP was enhanced during the early time in rat traumatic brain injury (TBI) model. The CHOP expression level of 24 h after TBI reached to the peak during the TBI period, and the expression amount was 11 times of the control.
