摘要
目的 研究CD147在子宫内膜异位细胞中的表达情况,并探讨其对子宫内膜细胞的迁移和凋亡的调节作用。方法 选取2013年6-12月于本院进行诊治的60例子宫内膜异位症患者为实验组,并以同期的60名不孕症患者为对照组,然后取实验组的异位内膜及对照组的正常子宫内膜进行检验,以实时荧光定量PCR法进行CD147的检测,统计并比较实验组进行抗体阻断前后的HES细胞迁移率及基质金属蛋白酶2(MMP-2)的表达。结果 实验组CD147表达为(7.65±1.25),其明显高于对照组(0.95±0.14),差异有统计学意义(P〈0.01);经抗体阻断后的HES细胞迁移率明显低于阻断前,差异有统计学意义(P〈0.01),阻断前后MMP-2表达则无明显差异(P〉0.05)。结论 CD147在子宫内膜异位细胞中呈现高表达状态,其通过MMP-2非依赖性途径来影响人子宫内膜上皮细胞HES的迁移,以达到参与疾病发生、发展的作用。
Objective To study the expression situation of CD147 in endometriosis,and investigate the regulating action of CD147 for the migration and apoptosis. Methods 60 patients with endometriosis in our hospital from June to December in 2013 were chosen as the experimental group,60 patients with infertility at the same time were chosen as the control group,then the ectopic endometrium of experimental group and normal endometrium of control group were detected,CD147 were detected with PCR real-time fluorescent quantitative method,and the HES cell migration rate and MMP-2 expression of experimental group were respectively analyzed and compared before and after the blocking antibodies. Results CD147 expression of experimental group was(7.65±1.25),and it was obviously higher than that of control group(0.95±0.14),with significant difference(P〈0.01),and the HES cell migration rate of experimental group after the blocking antibodies was obviously lower than that before the blocking antibodies with significant difference(P〈0.01),while the MMP-2 expression before and after the blocking antibodies had no obvious difference(P〉0.05). Conclusion CD147 in endometriosis shows high expression state,and it can influence the migration of human endometrial epithelial cell HES through the MMP-2 independent pathways,in order to participate in disease progression.
