摘要
[目的]筛选纤维素酶活力高的纤维素降解真菌,研究其粗酶活性。[方法]从土壤中分离、筛选高效纤维素降解菌,以透明圈试验和滤纸降解试验验证其降解能力,通过菌落菌丝形态及r DNA-ITS序列测序鉴定菌株种属,通过改变培养时间、氮源、装液量、起始p H及培养温度5个因素探讨纤维素降解真菌的最适产酶条件。[结果]得到3株纤维素降解真菌QS2、QS6和QW9,经鉴定确定QS2和QS6属青霉属(Penicillium),QW9为康宁木霉(Trichoderma koningii)。QS6菌株的FPA酶活和CMC酶活在3个菌株中均表现活力最高,且最适产酶条件为32℃时氮源为磷酸铵、起始p H 6.0、装液量100 ml,培养时间14 d。[结论]高效纤维素降解真菌粗酶活性的研究为纤维素酶的发酵生产提供一定的技术支持。
[ Objective] The research aimed to screen high efficiency cellulose degrading fungi and study their crude enzyme activity. [ Method ] The highly effective cellulose degrading fungi were screened from soil. The experiments of the transparent circle and degradation of filter paper were used to verify their degradation ability. The species were determined by their colony, mycelium morphology and rDNA-ITS sequence comparison. Five factors which included culture time, nitrogen source, liquid loading, starting pH and the culture temperature were used to determine the optimum conditions of cellulose degrading fungi. [ Result ] Three target strains were obtained, two of them were Penicilliurn named as QS2 and QS6, the other was Trichoderma koningii named as QW9. FPA activity and CMC activity of QS6 strain were the highest in the three strains. The QS6 strain performed the highest enzyme activity when it was cultured in 100 ml enzyme producing medium which nitrogen source was ammonium phosphate and the starting pH was 6.0 for 14 days. [ Conclusion] The research on the activity of the enzymes of the high efficiency cellulose degrading fungi provides a certain technical support for the production of cellulose.
