摘要
目的通过观察华蟾素注射液(Cmobutacini)对人胰腺癌细胞株SW1990、BxPC3细胞周期的影响以探讨其抑制肿瘤细胞增殖的作用机理。方法采用CCK-8法检测法在不同的时间点(12h、24h、48h、72h和96h)检测华蟾素注射液对人胰腺癌细胞株SW1990、BxPC3细胞增殖的影响;采用流式细胞术检测对细胞周期的影响。结果华蟾素注射液对人胰腺癌细胞株SW1990、BxPC3具有抑制细胞增殖的作用,并具有时间及剂量依赖性。其中对SW1990的半数抑制率浓度(IC50)为(8.92±1.98)mg/mL,对BxPC3的半数抑制率浓度(IC50)为(5.00±0.66)mg/mL。流式细胞术检测发现,华蟾素注射液可使人胰腺癌细胞株SW1990、BxPC3阻滞于的G2/M期。结论华蟾素注射液能通过诱导人胰腺癌细胞SW1990、BxPC3阻滞于G2/M期以抑制细胞增殖,为临床应用华蟾素注射液治疗胰腺癌提供实验依据。
Objective To investigate the effects of Huachansu on proliferation and cell cycle distribution of pancreatic cancer cell lines SW1990 and BxPC3 and effective mechanism. Methods The anti-proliferation effects of Huachansu on human pancreatic cell lines SW1990 and BxPC3 were tested by CCK-8 assay for 12 hours, 24hours, 48 hours, 72 hours and 96 hours, respectively. The cell cycles were analyzed with flow cytometry. Results The data of in vitro study showed there were a concentration and time dependent anti-proliferation effects of Huachansu on pancreatic cancer cell lines. The half maximal inhibitory concentrations (IC50) for 72 hours anti-proliferation of SW1990 and BxPC3 were ( 8.92 ± 1.98 ) mg/mL and (5.00±0.66) mg/mL, respectively. The cell cycle results showed that Huachansu could induce SW1990 and BxPC3 ceils arrest at G2/M phase. Conclusions Huachansu is able to inhibit the growth of pancreatic cell lines and induce cancer cells arrested at G2/M phase. Collectively, these findings indicate that G2/M phase arrest contributes to the anti-proliferation effects in pancreatic cancer therapy. Therefore, Huachansu may be used as a therapeutic agent for the treatment of pancreatic cancer.
