甘薯抗逆相关基因Rho的克隆和表达分析 被引量:2

摘要 Rho是小GTP结合蛋白家族的一个亚族,调控着肌动蛋白的重组过程,参与MAP激酶细胞信号转导过程。本试验通过RT-PCR从甘薯中克隆了1个Rho亚族成员IbRho,测序结果显示其含有504bp长的开放阅读框,编码167个氨基酸,推导出的分子量为18.43kDa。其与多种生物的Rho蛋白的氨基酸序列同源性为82.3%-94.0%,荧光定量PCR研究表明IbRho基因受盐、低温、PEG和植物激素ABA的诱导表达。IbRho基因的克隆为今后开展甘薯抗逆分子育种提供了重要的基因资源。 Rho protein is a member of small GTP-binding protein family controlling the recombination of actin protein and signal transduction of MAP kinase.In this experiment,Rho gene of sweet potato(IbRho)was amplified successfully by RT-PCR.Sequencing result showed that the cDNA of IbRho contained a continuous complete open reading frame(ORF)of 504 bp encoding apolypeptide of 167 amino acids with a calculated molecular mass of 18.43 kDa.The IbRho shared very high homology with many members of Rho subfamily from other species,with a 82.3%~94.0% homology.Treated with different kinds of environmental stimuli such as NaCl,cold,PEG and the plant hormone ABA,the transcription level of IbRhogene was differentially regulated using real-time PCR detection.IbRho gene may be an important gene resource for stress-resistant breeding of sweet potato.
