川芎含药血清对大鼠肝星状细胞大麻素受体1相关信号通路的影响 被引量:9

Effects of serum containing Ligusticum chuanxiong on CB1 related signaling pathway in rat hepatic stellate cells

摘要 目的观察川芎含药血清对血小板源生长因子(PDGF-BB)活化的大鼠肝星状细胞(HSCs)大麻素受体1(CB1)、黏着斑激酶(FAK)及磷酸化丝氨酸/苏氨酸蛋白激酶(p-Akt)蛋白表达的影响,探讨其抗纤维化的作用机制。方法制备川芎含药血清,MTT法检测川芎含药血清对肝星状细胞增殖情况,Western blot法分析肝星状细胞CB1、FAK、p-Akt蛋白的表达。结果与正常对照组相比,PDGF-BB作用24 h能刺激HSCs增殖,上调CB1、FAK、p-Akt蛋白的表达;川芎含药血清、秋水仙碱及大麻素受体1抑制剂AM251均能明显抑制上述效应;且加入AM251后,川芎含药血清对上述效应的抑制效果增强,且作用呈剂量依赖性。结论川芎含药血清可能通过大麻素相关信号通路抑制HSCs增殖,从而发挥防治肝纤维化的作用。 AIM To investigate the effects of serum containing Ligusticum chuanxiong on cannabinoid receptor1( CB1),focal adhesion kinase( FAK) and extracellular regulated protein kinase( p-Akt) in rat hepatic stellate cells( HSCs) stimulated by platelet-derived growth factor( PDGF-BB) and to explore the mechanism of hepatic fibrosis. METHODS The effect of prepared serum containing Ligusticum chuanxiong on the proliferation of HSCs was detected by MTT assay; the protein levels of CB1,FAK,and p-Akt were analyzed by Western blot. RESULTS Compared to the normal control group,24 hours treatment of PDGF-BB promoted the proliferation of HSCs significantly and increased the protein levels of CB1,FAK,and p-Akt; serums containing Ligusticum chuanxiong,colchicine and AM251 could distinctly inhibit the above effects in HSCs. After adding AM251,the inhibitory effect of serum containing Ligusticum chuanxiong was more apparent and showed a dose-dependent manner.CONCLUSION The serum containing Ligusticum chuanxiong plays the role in the prevention and treatment of hepatic fibrosis by inhibiting the proliferation of HSCs through CB1 related signaling pathway.
